Flotillin-1与Gab2蛋白在宫颈癌组织中的表达及与放疗敏感性的相关性

摘　　要：目的探讨Flotillin-1与Gab2蛋白在宫颈癌组织中的表达及与放疗敏感性的相关性。方法选取本院2014年2月～2016年2月收治的86例行根治性放疗的宫颈癌患者,根据近期疗效分为肿瘤消失组(54例)和肿瘤未控组(32例);根据远期疗效分为治愈组(50例)和复发组(36例)。收集其临床资料,行回顾性分析。通过Realtim-PCR技术对宫颈癌组织中Flotillin-1与Gab2蛋白表达进行检测,并比较不同根治性放疗敏感性患者宫颈癌组织中Flotillin-1与Gab2蛋白表达情况,再经Pearson相关性分析Flotillin-1与Gab2蛋白和放疗敏感性的相关性。结果放疗后Flotillin-1和Gab2 mRNA表达水平均低于放疗前(P<0.05);肿瘤消失组中的Flotillin-1mRNA、Gab2mRNA表达水平均低于肿瘤未控组(P<0.05);治愈组中的Flotillin-1mRNA、Gab2mRNA表达水平均低于复发组(P<0.05);肿瘤消失组宫颈癌组织中Flotillin-1mRNA、Gab2mRNA表达较低和肿瘤消失有显著正相关关系(r=0.455,P=0.022),肿瘤未控组宫颈癌组织中Flotillin-1mRNA、Gab2mRNA表达较高和肿瘤未控制有显著正相关关系(r=0.691,P=0.008);治愈组宫颈癌组织中Flotillin-1mRNA、Gab2mRNA表达较低和肿瘤治愈有显著正相关关系(r=0.457,P=0.021);复发组宫颈癌组织中Flotillin-1mRNA、Gab2mRNA表达较高和肿瘤复发有显著正相关关系(r=0.686,P=0.011);宫颈癌放疗敏感患者Flotillin-1mRNA、Gab2mRNA表达较低和放疗敏感有显著正相关关系(r=0.585,P=0.016),宫颈癌放疗抵抗患者中Flotillin-1mRNA、Gab2mRNA表达较高和放疗敏感有显著正相关关系(r=0.498,P=0.019)。结论 Flotillin-1与Gab2蛋白在宫颈癌组织中存在高表达,且Flotillin-1和Gab2蛋白高表达与放疗抵抗性有关。Objective To investigate the correlation of expression levels and radiosensitivity of Flotillin-1 and Gab2 proteins in cervical carcinoma.Methods The clinical data of 86 patients with cervical carcinoma treated at our hospital from February 2014 to February 2016 were retrospectively analyzed.The expression of the Flotillin-1 and Gab2 proteins in cervical carcinoma was detected by Realtim-PCR and compared among cervical carcinoma tissues with different radical chemotherapy sensitivity.Then the correlation of expression levels and radiosensitivity was analyzed by the Pearson correlation.Results The mRNA expression levels of Flotillin-1 and Gab2 after radiotherapy were lower than those before chemotherapy ( P <0.05).The expression level of Flotillin-1 mRNA and Gab2 mRNA in the tumor disappearance group were lower than those of the tumor disappearance group.The expressions of Flotillin-1 mRNA and Gab2 mRNA in tumor healing group were lower than those in tumor recurrence group.The low expression of Flotillin-1 mRNA and Gab2 mRNA in tumor disappearance group was positively correlated with tumor disappearance in cervical cancer tissues ( r =0.455,P =0.022).The high expression of Flotillin-1 mRNA and Gab2 mRNA in uncontrolled tumor group was positively correlated with uncontrolled tumors ( r =0.691,P =0.008).The low expression of Flotillin-1 mRNA and Gab2 mRNA in tumor healing group was positively correlated with tumor healing ( r =0.457,P =0.021).The high expression of Flotillin-1 mRNA and Gab2 mRNA in tumor recurrence group was significantly correlated with tumor recurrence ( r =0.686,P =0.011).The low expression of Flotillin-1 mRNA and Gab2 mRNA in cervical cancer radiotherapy-sensitive group was positively correlated with chemotherapy sensitivity ( r =0.585,P =0.016).The high expression of Flotillin-1 mRNA and Gab2 mRNA in the radioresistant group was positively correlated with chemotherapy sensitivity ( r =0.498,P =0.019).Conclusion Flotillin-1 and Gab2 proteins are highly expressed in cervical cancer tissues,which
