摘  要:【目的】确定肉苁蓉多糖的闪式提取工艺条件并分析其抗糖基化活性.【方法】采用单因素和响应面试验优化料液比、转速以及提取时间对多糖得率的影响,及其对赖氨酸/葡萄糖模拟体系中形成的晚期糖基化终产物(AGEs)的抑制效果.【结果】当料液比1∶53 (g/mL)、提取转速5 387 r/min、提取时间60 s时,多糖得率可达12.35%.制备多糖和阳性对照氨基胍(AG)对模拟体系中AGEs半数抑制浓度IC_(50)分别为0.438和0.341 mg/mL.【结论】采用闪式提取肉苁蓉多糖方法简单可行,回收率高,其多糖具有一定糖基化抑制效果.【Objective】To optimize the extraction process of polysaccharides from Cistanche deserticola(CDPs)using flash extraction,and to analyze antiglycosylation activity.【Method】The effect of different parameters ranging from solid-liquid ratio,voltage,and extraction time on the yield of CDPs were optimized through single factor experiments and response surface experiments.The effects of CDPs on the advanced glycation end products(AGEs)in Lysine/glucose models were also investigated.【Result】The optimal extraction conditions with a yield of 12.35% for extraction of CDPs were the ratio of solid to liquid of 1∶53(g/mL),a rotating speed of 5387 r/min,and the extraction time for 1 min.The IC50 values of CDPs and AG against AGEs were 0.438 and 0.341 mg/mL,respectively.【Conclusion】As a simple and efficient technique,it can be used to extract CDPs,which possess the inhibitory effects of antiglycosylation activity.
