摘 要:目的:分析多榔菊挥发油的化学成分,并初步考察多榔菊挥发油对DNA氧化损伤的保护作用。方法:采用水蒸气蒸馏法提取多榔菊挥发油,基于GC-MS法分析挥发油的化学成分,通过·OH诱导DNA氧化损伤并运用紫外-可见分光光度法测定多榔菊挥发油对DNA氧化损伤的保护作用。结果:检测到多榔菊挥发油中的20种化学成分,初步鉴定了12种化合物,占总挥发性成分97. 29%,其化学成分主要为2'-羟基-4'-甲氧基苯乙酮异丙醚(70. 26%)、4-(2-羟乙氧基)苯甲酸烯丙酯(18. 23%)、油酸(1. 98%)、棕榈酸乙酯(1. 67%)、肉豆蔻酸(1. 42%)、棕榈酸(1. 11%)、癸酸(0. 55%)和夹竹桃麻素(0. 38%)等;体外抗DNA氧化损伤试验表明,多榔菊挥发油具有抑制DNA氧化损伤作用,当浓度达到8μg·ml^(-1)时具有最佳保护作用。结论:结果表明多榔菊挥发油中主要含酮类,酯类和酸类成分,并具有较好的体外抑制DNA氧化损伤的能力。Objective:To analyze the chemical constituents of volatile oil from Doronicum hookeri L.and study its protective effect on DNA oxidative damage.Methods:The volatile oil was extracted from Doronicum hookeri L.by steam distillation method,the chemical components of volatile oil were analyzed by GC-MS,and the protective effect of volatile oil on·OH induced oxidative damage of DNA was determined by UV spectrophotometry.Results:Totally 20 kinds of chemical compositions were detected out from Doronicum hookeri L.volatile oil and 12 of them were identified preliminarily,which accounted for 97.29%of the total volatile components.The main chemical constituents were 2’-hydroxy-4’-methoxyacetophenone,isopropyl ether(70.26%),allyl p-(2-hydroxyethoxy)benzoate(18.23%),oleic acid(1.98%),hexadecanoic acid,ethyl ester(1.67%),tetradecanoic acid(1.42%),palmitic acid(1.11%),decanoic acid(0.55%)and apocynin(0.38%)etc.The in vitro anti DNA oxidative damage test showed that the volatile oil from Doronicum hookeri L.could suppress the DNA oxidation,and the best protective effect was observed at the concentration of 8μg·ml^-1.Conclusion:The volatile oil of Doronicum hookeri L.mainly contains ketones,esters and acids,and has better ability to inhibit oxidative damage of DNA in vitro.
