摘 要:目的研究叉头框蛋白M1(FoxMl)与中心体相关激酶2(Nek2)在肾癌组织中的表达及临床意义。方法随机选择肾癌患者25例,收集其术中切除的肾癌组织及其配对癌旁正常组织标本,标本均经病理检查确诊。采用免疫组织化学法和蛋白印迹法检测肾癌组织及其配对癌旁正常组织中FoxM1、Nek2蛋白的表达情况。结果肾癌组织中FoxM1蛋白相对表达量显著高于癌旁正常组织(P<0.05),肾癌组织中Nek2蛋白相对表达量也显著高于癌旁正常组织(P<0.05)。肾癌组织中FoxM1和Nek2的表达呈正相关(r=0.915,P<0.05)。结论肾癌组织FoxM1、Nek2表达均升高,二者可能协同促进肾癌的发生发展;癌组织FoxM1、Nek2表达变化可能有助于肾癌临床诊断和预后评估。Objective To study the expression change of forkhead box protein M1 (FoxM1) and NIMA-related kinase 2 (Nek2) in kidney cancer tissues and its clinical significance. Methods Twenty-five kidney cancer patients who underwent surgical removal of kidney cancer and adjacent tissues were randomly chosen. The specimens were diagnosed by pathology. The expression changes of FoxM1 and Nek2 were detected by Western blot and immunohistochemical method. Results The expression level of FoxM1 in kidney cancer tissues was significantly higher than that in adjacent tissues (P<0.05), the expression level of Nek2 in kidney cancer tissues was also significantly higher than that in adjacent tissues (P<0.05). The expression of FoxM1 and Nek2 in kidney cancer tissues was positively correlated (r=0.915, P<0.05). Conclusions The expression level of FoxM1 and Nek2 increases significantly in kidney cancer tissues, which may lead to the development and progression of kidney cancer. FoxM1 and Nek2 may be beneficial to the diagnosis and prognosis of kidney cancer.
