摘  要:目的探讨乌司他丁(UTI)对高转移人卵巢癌细胞HO-8910PM转移和侵袭能力的影响及其作用机制。方法培养HO-8910PM细胞,根据UTI剂量不同分为对照组及UTI低剂量组(400U/mL UTI)、中剂量组(800U/mL UTI)、高剂量组(1 600U/mL UTI),通过划痕实验、Transwel侵袭实验比较4组HO-8910PM细胞侵袭和转移能力;半定量RT-PCR结合Western blot法检测4组HO-8910PM细胞中尿激酶型纤溶酶原激活物(uPA)的表达水平。结果 4组HO-8910PM细胞迁移和侵袭能力比较,差异均有统计学意义(P<0.05)。随着UTI剂量增加,HO-8910PM细胞迁移距离和穿膜细胞数均呈明显减少趋势(P<0.05);uPA mRNA和蛋白表达水平呈明显降低趋势(P<0.05)。结论 UTI能够有效抑制人卵巢癌细胞HO-8910PM的侵袭和转移,且抑制作用存在剂量依赖性,其机制可能与UTI下调uPA的表达有关。Objective To investigate the inhibitory effect of Ulinastatin(UTI)on the invasion and metastasis in human ovarian carcinoma HO-8910 PM cell line.Methods HO-8910 PM cells were cultured and divided into 4 groups,the control group,the low-dose group,the medium-dose group and the high-dose group.The effect of UTI on HO-8910 PM cells on invasion and metastasis were analyzed by Matrigel invasion assay.RT-PCR assay and Western blot were used to determine the changes of mRNA and protein level of uPA.Results The various indexes differences in 4 groups of HO-8910 PM cells were statistically significant(P<0.05).The migration distance and the number of transmembrane cells showed a decreasing tendency(P<0.05).The mRNA and protein expression levels of uPA was significantly down-regulated(P<0.05).Conclusion UTI could inhibit the invasion and metastasis of the ovarian cancer cells.The inhibitory effect is dosedependent.The mechanism may be related to the down-regulation of uPA by UTI.
