摘 要:采用HPLC技术分析大豆疫霉抗病品种Williams82、感病品种Sloan及非寄主菜豆紫花油豆、玉米绥玉23种子分泌物中游离氨基酸和可溶性糖种类和含量,测定相对原位浓度氨基酸混合液和糖混合液对大豆疫霉菌丝生长、卵孢子形成及游动孢子趋化性的影响。结果表明,不同作物品种种子分泌物中氨基酸和糖的种类及含量差异显著。其中,大豆抗病品种种子分泌物中氨基酸和可溶性糖含量显著高于感病品种Sloan和其他两种非寄主作物。对于菌丝生长,除Williams82氨基酸处理表现为抑制外,其他处理均有促进。对于卵孢子形成,氨基酸处理表现为紫花油豆=Williams82>Sloan=绥玉23,可溶性糖处理表现为紫花油豆>Sloan>Williams82>绥玉23。对于游动孢子趋化性,表现为Sloan>紫花油豆>Williams82>绥玉23。4个品种种子分泌物差异主要表现在对游动孢子差异性趋化作用上,表明种子分泌物中游离氨基酸和可溶性糖在一定程度上作为信号分子参与大豆疫霉寄主选择性和寄主抗病性,研究为防治大豆疫霉根腐病提供理论依据。The free amino acids and soluble sugars in the seed exudates of soybean resistant varieties Williams 82, susceptible varieties Sloan and non- hosted common bean Zihuayoudou, maize Suiyu 23 were analyzed by High Performance Liquid Chromatography(HPLC). The effects of relative in situ concentration of free amino acid mixture and soluble sugar mixture on mycelial growth, oospores formation and zoospores chemotaxis of Phytophthora sojae were determined by cultivars. The results showed that there were significant differences in the kinds and contents of amino acids and sugars in seed exudates of different cultivars. Among them, the contents of amino acids and sugars in the exudates of soybean resistant cultivar were significantly higher than those of other three cultivars. For mycelial growth, except for Williams 82 amino acid treatment showed inhibition, other treatments promoted mycelial growth. For oospores formation, the amino acid treatment was represented as Zihuayoudou=Williams 82>Sloan=suiyu 23, and the sugar treatment was represented as Zihuayoudou> Sloan>Williams 82>suiyu 23. For zoospore chemotaxis, they were shown as Sloan>Zihuayoudou> Williams 82> suiyu 23. The differences of seed exudates of the four cultivars were mainly reflected in the differential chemotaxis to zoospores, it indicated that amino acids and sugars could be a signal molecule in seed exudates and participated in host selectivity and host resistance of Phytophthora sojae, the results provided a theoretical basis for the use of non- host and host disease resistance to control Phytophthora root and stem rot.
