摘  要:目的:探讨原代哺乳期乳腺细胞中促甲状腺激素(TSH)对钠碘同向转运体(sodium iodide symporter,NIS)的调节作用。方法:在不同浓度TSH 作用下培养小鼠原代哺乳期乳腺细胞,采用实时荧光定量PCR 和Western blot 法检测NIS mRNA和蛋白的表达;Western blot法检测沉默细胞的促甲状腺激素受体(TSHR)后NIS蛋白的表达以及抑制细胞的蛋白激酶A(PKA)后NIS蛋白的表达。结果:小鼠原代哺乳期乳腺细胞TSH高剂量组NIS mRNA表达量最高(P<0.05),NIS蛋白表达差异无统计学意义(P>0.05);沉默TSHR后各组NIS蛋白表达之间差异无统计学意义(P>0.05);PKA抑制组NIS蛋白表达低于TSH组和空白组(P<0.05)。结论:高浓度TSH对小鼠哺乳期乳腺细胞NIS表达具有一定调节作用;PKA可上调NIS的表达。Objective:To study the regulation mechanism of thyroid stimulating hormone(TSH)to sodium iodide symporter (NIS)in original generation lactating mammary cells of mice. Methods: Original generation lactating mammary cells of mice was cultured under different concentrations of TSH. The expression of NIS mRNA and protein were detected by real-time quantitative PCR and Western blot.The expression of NIS protein after silencing of TSHR of primary cells and after inhibiting protein kinase A (PKA)of primary cells were detected. Results: The highest expression level of NIS mRNA was found in high dose TSH group of original generation lactating mammary cells of mice(P<0.05), and there was no significance in the expression of NIS protein.The expression of NIS protein after inhibiting PKA was lower than that in TSH group and blank group(P<0.05). There was no significant difference in the expression of NIS protein after silencing of TSHR of primary cells(P>0.05). Conclusion: The high concentration of TSH can increase the expression of NIS in original generation lactating mammary cells and PKA plays an important role in increasing the expression of NIS.
