摘 要:目的探讨不同的洋地黄类药物对甲状腺乳头状癌细胞株BC-PAP的自噬调控作用。方法将甲状腺乳头状癌细胞株BC-PAP分为空白对照组、洋地黄毒苷组、地高辛组,应用CCK-8方法测定细胞活性,并观察细胞的生长情况,电镜观察细胞内自噬小体的形成。结果洋地黄毒苷、地高辛均能够呈剂量依赖性地抑制BC-PAP细胞的生长。洋地黄毒苷的IC50浓度为0.25μmol/L,而地高辛的IC50浓度为5μmol/L。结论洋地黄毒苷、地高辛可增强甲状腺癌细胞自噬,洋地黄毒苷诱自噬的作用更强。Objective To investigate digitalis-like compounds facilitating autophagy in papillary thyroid carcinoma. Methods BC-PAP cells were divided into three groups:control,digitoxin and digoxin group. The half-inhibition rate of the cells was calculated by cck-8 experiment. The grouth condition of cells was abserved cells of each group were cultured for 12 hand observed under transmission electron microscope. The formation of autophagosomes in cells was observed under electron microscope. Results Under electron microscope, the formation of autophagosomes in digitoxin group and digoxin group were significantly higher than those in the blank control group. IC50 of digitoxin was 0.25 μmol/L. IC50 of digoxin was 5 μmol/L. Conclusion Digitalis-like compounds can increase autophagy in papillary thyroid carcinoma. Digitoxin was more powerful than digoxin.
