摘 要:目的:探讨p15^(PAF)蛋白对食管癌细胞糖酵解水平及相关生物学效应的影响。方法:使用细胞活性检测及Western Blot等方法对细胞耗糖量、2-脱氧葡萄糖(2-DG)的敏感性及自噬水平等生物学效应进行检测。结果:上调p15^(PAF)蛋白的食管癌Eca-109及TE-1细胞较对照组有更高的增殖能力和葡萄糖消耗,但对2-DG造成的细胞增殖抑制更为敏感,下调p15^(PAF)蛋白的细胞细胞增殖能力下降,该现象与Akt信号通路有关。另外,p15^(PAF)蛋白还可以通过Akt信号通路下调食管癌细胞的基础自噬水平。结论:p15^(PAF)蛋白可以通过PI3K-Akt信号通路影响食管癌细胞的糖酵解及自噬水平。Objective:To discover the effect of p15^PAF protein on glycolysis of esophageal cancer cells.Methods:We used MTT test to detect cell glucose comsumption,and sensitivity of 2-dexyo-glucoseand we performed Western Blot of LC-ⅢB to analyze the level of autophagy,HK-2 and GLUT-1 to assess the glycolysis.Results:Elevated p15^PAF protein increased proliferation ability and glucose comsumption,while significantly inhibited when incubating with 2-DG.Down regulation of p15^PAF caused the opposite result,and this phenomenon related with Akt signaling.On the other hand,p15^PAF protein decreased basic autophagy ability via Akt signaling with the mechanism needing studied.Conclusion:p15^PAF protein may increase glycolysis ability and decrease basic autophagy by activating PI3 K-Akt signaling.
