摘 要:目的通过分析癌症基因组图谱(the Cancer Genome Atlas,TCGA)数据库中的胶质母细胞瘤(glioblastoma multiforme,GBM)患者的测序数据以及本院GBM患者临床样本,评估GBM中表皮生长因子(epidermal growth factor receptor,EGFR)的mRNA表达情况及其与患者预后的关系。方法分析TCGA数据库中520例GBM患者的测序及临床数据,然后通过实时定量PCR(real-time polymerase chain reaction,RT-PCR)在本院42例GBM样本、2例Ⅰ级胶质瘤和4例对照脑组织样本中进行验证。结果 TCGA数据库数据分析发现mRNA高表达组中有大量错义突变,本院临床样本胶质母细胞瘤中EGFR的mRNA表达水平明显升高。Kaplan-Meier生存曲线分析结果表明,TCGA数据库中EGFR高表达组的GBM患者的总体生存期显著低于非EGFR高表达组(P=0.003 64),而无进展生存期差异无统计学意义(P=0.163)。本院患者中这两组的总体生存期差异无统计学意义。结论 EGFR可能作为一种GBM患者预后的潜在预测因子,对胶质瘤的诊断和治疗起到关键性作用。Objective To analyze the association between epidermal growth factor receptor(EGFR) mRNA expression and the prognosis of glioblastoma multiforme(GBM) patients. Methods The mRNA expression profiles and clinical data of 520 GBM patients in TCGA database were analyzed. And 42 glioblastoma, 2 Grade I glioma and 4 normal brain tissue samples collected from Shenzhen Second People′s Hospital were also analyzed. Results TCGA data analysis showed that there were a large number of missense mutations in GBM patients with high mRNA expression;and the mRNA expression level of EGFR in the our clinical samples was significantly increased. Kaplan-Meier survival analysis showed that the overall survival of GBM patients in the TCGA database with high EGFR expression was significantly lower than that of the non-EGFR-high expression patients(P=0.003 64), while there was no significant difference in the progression-free survival(P=0.163). However, there was no significant difference in overall survival between the two groups of patients in our hospital. Conclusion EGFR may be a potential prognostic factor of GBM patients and play a key role in the diagnosis and treatment of glioma.
