摘  要:目的探讨5-氟尿嘧啶(5-FU)缓释植入制剂--氟尿嘧啶植入剂,对宫颈癌Hela细胞增殖和凋亡的影响。方法将加入5-FU和氟尿嘧啶植入剂的Hela细胞分别作为5-FU组和氟尿嘧啶组,未作处理的细胞作为对照组。CCK8法检测细胞增殖能力,流式细胞仪检测细胞凋亡情况,蛋白质印迹法(Western blot)检测B细胞淋巴瘤/白血病-2(Bcl-2)和caspase 3蛋白的相对表达量。结果干预2、3、5、7天时,5-FU组细胞OD值均低于对照组细胞(P﹤0.05),但仅干预5、7天时,氟尿嘧啶组细胞OD值均低于对照组细胞(P﹤0.05)。干预3天时,氟尿嘧啶组细胞凋亡率低于5-FU组细胞和对照组细胞,5-FU组细胞凋亡率高于对照组细胞(P﹤0.05);干预7天时,氟尿嘧啶组和5-FU组细胞凋亡率均高于对照组细胞(P﹤0.05)。干预3、7天,5-FU组和氟尿嘧啶组细胞Bcl-2蛋白相对表达量均低于对照组细胞,caspase 3蛋白相对表达量均高于对照组细胞,差异均有统计学意义(P﹤0.05)。结论氟尿嘧啶植入剂可能通过上调caspase 3的表达,下调Bcl-2的表达,从而抑制宫颈癌Hela细胞的增殖,与促进细胞凋亡有关。Objective To explore the effects of 5-fluorouracil(5-FU)sustained--release implant preparation-fluorouracil implants on the proliferation and apoptosis of cervical cancer cells.Method Hela cells treated with 5-FU and fluorouracil implants were assigned as 5-FU group and fluorouracil implants group,and untreated cells were included as control.The proliferative activity of Hela cells was detected by CCK8,and cell apoptosis was determined by flow cytometry,the relative expression of B cell lymphoma/leukemia-2(Bcl-2)and caspase 3 protein were detected by Western blot.RRee-sult On the 2nd,3rd,5th,and 7th day of treatment,the optical density(OD)value in 5-FU group were consistently lower compared to control group(P<0.05),while on 5 thand 7 thday of treatment,the OD value in fluorouracil implants group was also lower in comparison with that in control group(P<0.05).On 3rd day of treatment,the cell apoptosis rate in fluorouracil implants group was significantly lower compared to 5-FU group and control group,and it was higher in 5-FU group than in control group(P<0.05);on 7th day of treatment,the cell apoptosis rate in fluorouracil implants group and 5-FU group were all higher than that in control group(P<0.05).On 3rd and 7th day of treatment,the relative expression of Bcl-2 proteins was lower in 5-FU group and fluorouracil implants group than that in control group,while the relative expression of caspase 3 protein was higher than that in control group,indicating statistically significant difference(P<0.05).Conclusion The possible explanation is,by upregulating the expression of caspase 3 and downregulating Bcl-2 expression,fluorouracil implants inhibits the proliferation of cervical cancer Hela cells,which is associated with the promotion of cell apoptosis.
