摘 要:目的:研究甲状旁腺激素(PTH)对下颌升支截骨术后正畸牙移动过程中骨保护素(OPG)和核因子κB受体活化因子配体(RANKL)的表达。方法:48只兔建立下颌升支截骨和下颌第一磨牙正畸移动模型,随机分成2组。实验组术后间歇性注射PTH 20μg/kg,对照组注射生理盐水。免疫组化方法和荧光定量PCR检测正畸牙牙周组织中RANKL和OPG表达变化。结果:实验组正畸牙压力侧牙周组织RANKL蛋白及mRNA表达大于对照组(P<0.05),而OPG蛋白及mRNA表达均小于对照组(P<0.05)。结论:外源性PTH可通过促进正畸牙压力侧牙周组织中RANKL表达,抑制OPG的表达,加速下颌升支截骨术后正畸牙的移动。Objective: To investigate the expression of osteoprotegerin(OPG) and receptor activator of nuclear factor-κB ligand(RANKL) in orthodontic tooth movement after mandibular ramus osteotomy regulated by parathyroid hormone(PTH). Methods: An orthodontic tooth movement model of mandibular first molars after mandibular ramus osteotomy was established in 48 rabbits, then the rabbits were randomly divided into 2 groups. The rabbits in experimental group were administered intermittent subcutaneous injection of 20 μg/kg PTH, those in the control group were given normal saline. RANKL and OPG expression in the periodontal tissues of the molars was examined by immunohistochemistry and q-PCR. Results: Experimental group exhibited significantly higher RANKL mRNA level and lower OPG mRNA level on the compression side of periodontal tissues than the control group. Conclusion: Administration of PTH can accelerate the movement of orthodontic tooth after mandibular ramus osteotomy by promoting the expression of RANKL and inhibiting the expression of OPG in periodontal tissue.
