摘  要:目的观察慢性束缚应激(chronic restraint stress,CRS)对小鼠行为学的影响及海马锌指蛋白A20变化,为阐明应激致抑郁的发病机制提供实验依据。方法通过CRS建立应激致抑郁动物模型,8w后测定动物体质量,并检测小鼠悬尾不动时间、自主活动运动速度、糖水偏爱分数等行为学指标;ELISA法检测海马组织TNF-α和IL-6含量;实时荧光定量PCR法和蛋白印迹检测海马A20 mRNA和蛋白表达水平。结果与对照组相比,8w CRS暴露后,小鼠体质量明显降低;自发活动的平均速度和糖水偏爱分数明显降低,而悬尾不动时间显著延长;小鼠海马组织内TNF-α和IL-6的含量显著增加;海马组织内A20mRNA和蛋白的表达量显著降低。结论 CRS导致小鼠抑郁样行为学改变可能与神经炎症和A20表达降低相关。Objective To investigate the effects of chronic restraint stress(CRS) on the behavior of mice and the change of zinc finger A20 in hippocampus. Methods The body weight of mice was measured after 8 weeks of CRS exposure. Chronic stress model was proved to be successfully established by the movement speed of spontaneous activity test,immobility time of tail suspension test and sucrose preference test. TNF-α and IL-6 contents in hippocampus were measured by ELISA. Meanwhile, real time PCR and Western blot were performed to determine the mRNA and protein expression levels of A20 in the hippocampus of CRS-induced mice. Results It was shown that the body weight, movement speed of spontaneous activity and sucrose preference decreased significantly in CRS group. Meanwhile, CRS exposure significantly increased the immobility time of tail suspension test. TNF-α and IL-6 contents increased and A20 mRNA and protein expressions decreased in hippocampus after CRS exposure compared to the control. Conclusion Hippocampal neuroinflammation and decreased A20 expression may play a role in the development of CRS-induced depression-like behavior in mice.
