摘 要:目的 利用Oncomine基因数据库分析CRABPⅡ基因在乳腺癌中的表达及对乳腺癌患者生存的意义。方法 通过Oncomine、Kaplan-Meier Plotter和癌症细胞系百科全书分析CRABPⅡ基因在各类乳腺癌组织中的表达和基因差异表达,并进行Meta分析和乳腺癌患者预后生存分析。结果 Oncomine数据库检索CRABPⅡ基因有438项不同种类结果。50项研究的表达有统计学差异,其中39项研究显示表达增高,11项研究显示表达降低,共有研究样本量653例;癌症细胞系百科全书显示CRABPⅡ基因在乳腺癌细胞系中的表达水平最明显;Kaplan-Meier Plotter数据库检索结果表明CRABPⅡ高表达导致患者的总生存预后不良( P =0.0035);与低表达组相比,CRABPⅡ高表达组乳腺癌患者的总生存时间显著降低( P <0.05);同时,CRABPⅡ的高表达也是luminal A型乳腺癌预后不良的重要因素。结论 通过深入挖掘Oncomine等数据库中CRABPⅡ基因芯片信息,证实CRABPⅡ在乳腺癌组织与细胞中高表达,且与乳腺癌患者生存预后不良相关,为临床乳腺癌的治疗及基因靶向药物的研制提供重要依据。Objective To analyze the expression of CRABPⅡ gene in breast cancer using Oncomine gene database and its significance to the survival of breast cancer patients. Methods By using Oncomine,Kaplan-meier Plotter,and Cancer Cell Line Encyclopedia,CRABPⅡ gene expression and gene differential expression in various cancer tissues were analyzed,and meta-analysis,prognosis and survival analysis of breast cancer patients,and breast cancer cell line analysis were conducted. Results Oncomine database retrieval CRABPⅡ gene 438 different species as a result,the expression of 50 studies have statistically significance,among them 39 study showed higher expression,11 study showed reduced expression,study sample size 653 cases.CRABPⅡ gene was retrieved by Cancer Cell Line Encyclopedia,indicating high expression in breast cancer tissue.The Kaplan-Meier Plotter database search results showed that the higher expression of CRABPⅡ had positive relationship with poor overall survival( P =0.0035).Compared with the low expression group,the total survival time of breast cancer patients with high expression of CRABPⅡ was significantly lower( P <0.05).At the same time,the high expression of CRABPⅡ was also an important factor in the poor prognosis of luminal A breast cancer. Conclusion By using in-depth digging Oncomine CRABPⅡ gene chip information in the database,such as confirmed CRABPⅡ organization high expression in breast cancer,and significantly associated with the prognosis of patients with breast cancer survival,for the clinical treatment of breast cancer and gene provide important basis for the development of targeted drugs.
