摘  要:为了研究花生四稀酸(AA)对绒山羊毛乳头细胞(DPCs)增殖的影响,探讨AA对DPCs中维生素D受体(VDR)基因表达的影响机制。通过体外培养DPCs给予不同浓度的AA作用不同的时间,采用CCK8法检测DPCs存活率,利用免疫荧光染色与实时定量PCR检测VDR的表达水平变化。通过体外培养DPCs和VDR基因缺陷型DPCs给予AA处理,利用实时定量PCR检测毛囊相关基因(IGF1、Noggin、b-catenin和Lef1)的mRNA表达变化。结果表明:AA(10-7mol/L)可显著促进DPCs的增殖(P<0.05),显著提高VDR基因的mRNA和蛋白表达;显著提高IGF1和Lef1基因的mRNA表达(P<0.05);VDR基因缺陷型DPCs中添加AA时,IGF1基因的mRNA表达没有显著变化。研究结果表明,AA促进DPCs细胞的增殖和毛囊中VDR、IGF1和Lef1基因的表达,且AA对于毛囊中IGF-1表达的促进作用受到VDR基因的调控作用。To investigate the influence of arachidonic acid(AA)on the proliferation of dermal papilla cells(DPCs)in cashmere goats and seek the mechanism of AA on the expression of vitamin D receptor gene(VDR),DPCs were cultured in vitro and treated with different concentrations of AA for different periods.The viability of DPCs survival rate was detected by CCK8,the expression level of VDR was detected by immunofluorescence staining and RT-PCR,the mRNA expression level of hair follicle cycle related genes(IGF1,Noggin,b-catenin and Lef1)were detected by RT-PCR.Results showed the treatment with 10-7 M of AA could significantly increase the the activity of DPCs,and significantly increase the expression of VDR mRNA and fluorescent protein in DPCs(P<0.05).AA significantly increased the expression of IGF1 and Lef1 genes in DPCs(P<0.05),while IGF1 was not affected by AA in VDR gene-deficient DPCs.This study proved AA could promote the proliferation of DPCs and the expression of VDR,IGF1 and Lef1.The effect of AA on the expression of IGF1 in hair follicle is regulated by VDR.This research will provide new ideas for the regulation of hair follicle formation and development of cashmere goats.
