摘  要:目的:探讨miR-337-3p对胃癌细胞SGC7901中HPSE表达的调控作用及其对细胞增殖的影响。方法:miR-337-3p mimics转染SGC7901细胞后,应用RT-PCR、Western blot检测其对miR-337-3p表达水平的影响及其对HPSE表达的影响;荧光素酶实验检测转染细胞中HPSE 3’-UTR荧光素酶活性;CCK-8测定细胞增殖的影响。结果:上调miR-337-3p可以抑制HPSE的表达、降低HPSE 3’-UTR启动子荧光素酶活性,并可抑制胃癌细胞增殖。结论:miR-337-3p可以通过下调HPSE的表达抑制胃癌细胞SGC7901增殖。Objective: To explore the mechanism of miR-337-3p regulating the expression of HPSE and the effect of proliferation of gastric cancer cells SGC7901. Methods: Mi R-337-3p mimics were transfected into SGC7901 cells,expression of miR-337-3p and HPSE were detected by RT-PCR and Western blot.Dual luciferase reporter assay was used to detect the target relationship between miR-337-3p and HPSE 3’-UTR promoter.Then cell proliferation was detected by CCK-8 assay. Results: Exogenous expression of miR-337-3p could inhibit the expression of HPSE and luciferase activity of HPSE 3’-UTR promoter.Upregulation of miR-337-3p also inhibited cell proliferation of SGC7901 cells. Conclusion: Mi R-337-3p is potentially involved in proliferation in gastric cancer cells SGC7901 via its regulating the expression of HPSE.
