摘 要:目的利用蛋白芯片筛选高血压脑梗死患者血浆中可能用于脑梗死早期诊断、预后判断的血浆标志物:方法采用生物素标记抗体芯片技术,对12名高血压患者及12名由高血压引起的脑梗死(高血压脑梗死)患者进行血浆蛋白质组学的检测。采用芯片分析软件提取数据,获得的数据采用AAH-BLG-1000的数据分析软件作统计分析:结果高血压脑梗死组和高血压对照组相比,生长因子M-CSF、Progranulin、PD-ECGF、VEGF-C,细胞因子TRANCE、TWEAK,具有抗氧化活性的结合珠蛋白Haptoglobin(HP)、过氧化物酶PRDX6,具有信号转导作用的TNF受体超家族成员10C(TNFRSF10C)、DKK类似蛋白1(Soggy-1)、Tool样受体3(TLR3)、Fibrinogen、IL-17RD,趋化因子系统的CCR4、CXCL16、白细胞衍生趋化因子(LECT2).簇集蛋白Clusterin,基质金属蛋白酶MMP-19、MMP-20,以及巢蛋白(Nidogen-1)共20个血浆蛋白差异表达(P<0.05,Foldchange绝对值>1.5)。结论蛋白质芯片共检测出20个潜在的高血压脑梗死血浆标志物,这些血浆蛋白参与了脑梗死发生发展过程中的多条路径,如细胞趋化、黏附、迁移、增殖,血栓形成和血管再生,以及介导炎症和免疫反应。蛋白芯片有助于早期鉴别脑卒中类型,诊断脑梗死及判断患者的预后,指导更有效更准确的临床治疗。Objective To screen plasma biomarkers in patients with hypertensive cerebral infarction using biotin-labeled protein chips that can be applied to hypertensive cerebral infarction in its early diagnosis, conditions and prognosis. Methods Biotin-labeled protein chips were used to detect 24 plasma including 12 hypertensive cerebral infarction and 12 hypertension controls. Using Microarray analysis software was used to extract the data, the results obtained were analyzed by AAH-BLG-1 data analysis software. Results Compared with the hypertension group, growth factor including M-CSF, Progranulin, PD-ECGF, VEGF-C, and Cytokine including TRANCE, TWEAK/TNFSF12, anti-oxidation including Haptoglobin, PRDX6, Signal Transduction including TNFRSF10 C, Soggy-1, TLR3, Fibrinogen, IL-17 RD, chemokine including CCR4,CXCL16,LECT2, matrix metalloproteinase including MMP-19,MMP-20 and Clusterin, Nidogen-1 were all expressed differently(P<0.05,Foldchange absolute value 1.5). Conclusion A total of 20 plasm proteins in patients with hypertensive cerebral infarction were screened using biotin-labeled protein chips, they participate in the hypertensive cerebral infarction cell chemotaxis, adhesion, migration, proliferation, thrombosis and angiogenesis, mediating inflammation and immune responses. Protein microarray can be helpful to identify early stroke types, diagnose cerebral infarction and have a better judgment of the patients' prognosis as well as guiding effective and accurate clinical treatment.
