摘 要:目的检测CARD10在乳腺癌组织中的表达情况及体外沉默其表达后对MDA-MB-231乳腺癌细胞周期、增殖能力的影响。方法利用免疫组织化学染色检测CARD10在62例乳腺癌及8例正常乳腺组织中的表达;利用化学合成的靶向CARD10-siRNA转染MDA-MB-231, 48 h后,RT-PCR检测干扰效率,流式细胞实验检测细胞周期变化,CCK-8检测增殖能力变化。结果CARD10在乳腺癌组织中显著性高表达,且与乳腺癌患者分期相关(P<0.05);CARD10被沉默后,抑制M DA-MB-231细胞增殖,阻滞细胞周期进程。结论CARD10促进乳腺癌MDA-MB-231细胞周期进程和细胞增殖,该研究为CARD10在乳腺癌的靶向治疗奠定了基础理论依据。Objective This study examined the expression of CARD10 in breast cancer and the effect of silencing its expression in vitro on cell cycle and proliferation of MDA-MB-231 breast cancer cells. Methods The expression of CARD10 in 62 cases of breast cancer and 8 cases of normal breast tissue was detected by immunohistochemical staining. MDA-MB-231 was transfected with chemically synthesized targeted CARD10-siRNA. After 48 hours, interference efficiency was detected by RT-PCR, cell cycle was detected by flow cytometry, and proliferation ability was detected by CCK-8. Results The expression of CARD10 was significantly higher in breast cancer tissues and correlated with the stage of breast cancer (P <0. 05 ). After the silence of CARD10, the proliferation of MDA-MB-231 cells was inhibited and the cells were arrested in the cell cycle. Conclusion CARD10 promotes the cell cycle and proliferation of breast cancer MDA-MB-231. This study lays a theoretical foundation for the targeted treatment of breast cancer with CARD10.
