摘  要:SET作为多功能的调控因子,涉及DNA复制转录调控、组蛋白及染色体修饰、细胞增殖凋亡等多种生物过程。SET蛋白广泛存在于各组织器官,调控多脏器多系统生理过程及疾病的发生、发展。SET的表达和活性与癌症的发生、转移及预后相关。SET通过影响组蛋白乙酰化修饰、DNA甲基化、染色质重塑等调控基因转录导致肿瘤发生。SET表达异常可导致DNA损伤修复异常最终导致细胞癌变或死亡。同时SET还调控相关因子磷酸化表达影响细胞迁移、细胞周期、细胞凋亡等,导致肿瘤的发生、发展。因此,SET蛋白被认为是癌症治疗的潜在靶点,目前正在开发用于临床治疗的抑制剂。(Patient SE translocation)SET is a multifunctional regulatory protein, involving various biological processes such as DNA replication and transcriptional regulation,histone and chromosome modification,cell proliferation and apoptosis.SET protein widely exists in various tissues and organs and regulates the occurrence and development of multi-organ and multi-system diseases.The expression and activity of SET are related to the occurrence, metastasis and prognosis of cancer.SET regulates gene transcription and induces tumorigenesis by affecting histone acetylation,DNA methylation,chromatin remodeling and other aspects.Abnormal expression of SET can lead to abnormal repair of DNA damage and eventually lead to cancer or death of cells.SET also regulates the phosphorylation of related factors,affecting cell migration,cell cycle and apoptosis,and affecting the occurrence and development of tumor.Therefore, SET protein is considered as a potential target for cancer treatment,and several SET inhibitors are currently being developed for the clinical treatment.
