摘 要:目的:研究化疗药物阿霉素体外诱导食管癌细胞衰老的时间和形态学特点。方法:不同剂量的阿霉素按两种方案处理食管癌细胞EC9706,SA-β-Gal染色表征衰老细胞,RT-PCR检测衰老相关分泌因子的表达。细胞图像分析比较SA-β-Gal阳性和阴性细胞间的形态差异。结果:不同剂量阿霉素处理EC9706细胞3 d后SA-β-Gal染色阳性细胞比例较低,经过在无药物培养基中延时培养6 d后可显著提高SA-β-Gal染色阳性率。RT-PCR结果显示延时培养能进一步增强衰老相关分泌因子的表达。衰老细胞中,上皮样形态细胞比例低于间质样细胞。结论:阿霉素诱导的食管癌细胞衰老具有时滞效应,上皮-间质转化可抑制细胞衰老。Objective To investigate the temporal and morphologic characteristics of esophageal cancer cell senescence induced by adriamycin in vivo. Methods EC9706 esophageal cancer cells were treated with different doses of adriamycin according to two schemes. Cell senescence was detected by SA-β-Gal staining. Senescence associated secretary factors were detected by RT-PCR. Cell image analysis was used to compare the morphology between SA-β-Gal positive and negative cells. Results The percentage of SA-beta-Gal positive cells was low after 3 days of adriamycin treatment. The positive rate of SA-beta-Gal staining was significantly increased after 6 days of delay culture in drug-free medium. RT-PCR results showed that delayed culture could further enhance the expression of senescence-related secretory factors. The proportion of epithelial-like cells in senescent cells was lower than that of mesenchymal-like cells. Conclusion Adriamycin-incuded senescence of esophageal cancer cells has a delayed effect,and epithelial-mesenchymal transition could inhibit cell senescence.
