摘 要:毛囊是皮肤的附属器官之一。有研究表明外胚层发育不全基因(ectodysplasinA,EDA)是毛囊、牙齿和腺体等器官发育所必需的,EDA及其受体EDAR可以参与并调节毛囊不同时期的发育。[目的]探究EDA和EDAR对小鼠背部毛囊形成的作用。[方法]试验采用苏木精-伊红(HE)染色和免疫组织化学方法,观察小鼠胚胎期第13~18天背部毛囊形成过程中组织结构特征以及EDA和EDAR蛋白在此期间的表达。[结果]HE染色试验结果显示不同天数胚胎小鼠背部毛囊结构有如下变化:细胞排列逐步紧密,层数逐渐增多;皮肤毛胚逐渐凸起,形成基板,并逐步发育成毛钉,进而形成未成熟毛囊。免疫组织化学试验结果显示EDA和EDAR蛋白在表层细胞、基底层、基板、毛芽、毛钉、未成熟毛囊和真皮中均有表达。[结论]EDA可能对毛囊形成的启动有促进作用,且可能促进毛钉的形成和未成熟毛囊的发育;EDAR则可能对表皮的分化有诱导作用,且可能对毛钉的形成和发育有促进作用。Hair follicle is one of the skin appendages. Studies have shown that ectodysplasin A (EDA), an ectodermal dysplasia gene, is necessary for the development of hair follicles, teeth, and glands. EDA and its receptor EDAR can participate in and regulate the development of hair follicles at different developmental stages.[Objectives] Present study was conductedto investigate the effects of EDA and EDAR on the formation of dorsal hair follicles of mice at embryonic stage.[Methods] Hematoxylin-eosin (HE) staining and immunohistochemistry were used in this study to observe the tissue structure characteristics and the expression of EDA and EDAR proteins during the formation of dorsal hair follicles between 13th and 18th day of embryonic period of mice.[Results]The results of HE staining test showed that the structures of dorsal hair follicles of embryonic mice at different days were changed in the way that the cells were gradually compressed and the number of layers were increased gently;Hair germs were bulged steadily and formed placode which developed to hair peg later on, and eventually formed immature hair follicles.Immunohistochemistry test results indicated that EDA and EDAR proteins were expressed in epidermis cells, basal layers, placodes, hair buds, hair pegs, immature hair follicles, and dermis.[Conclusions] EDA may promote the initiation of hair follicles formation, and contribute to the formation of hair pegs and the development of immature hair follicles;EDAR may induce the differentiation of epidermis and stimulate the formation and development of hair pegs.
