小鼠白假丝酵母菌阴道炎模型的优化及其效果评价

摘　　要：目的构建经济实用的白假丝酵母菌阴道炎小鼠模型。方法采用ICR雌性小鼠,预先给予低、高剂量(0.015,0.15mg/10g)的戊酸雌二醇连续灌胃7d,注射地塞米松0.15mg免疫抑制后接种白假丝酵母菌建立阴道炎模型,并进行阴道灌洗液菌落计数和阴道组织病理观察。结果戊酸雌二醇低、高剂量组阴道灌洗液的菌落计数在第4天达到峰值,分别为3.324±0.150,6.825±1.247,差异比较有统计学意义(P<0.05),注射地塞米松后戊酸雌二醇低、高剂量组阴道灌洗液的菌落计数在第7天达到峰值分别为8.472±0.954,14.152±3.24,比较差异有统计学意义(P<0.01)。病理HE染色后显示,高剂量戊酸雌二醇+地塞米松组有较多多形核粒细胞(PMNs)浸润。结论地塞米松+高剂量戊酸雌二醇为ICR小鼠白假丝酵母菌阴道炎模型最佳造模条件。并在一周时达最佳状态。Objective To establish Candida albicans vaginitis in mice and evaluate its effection. Methods Female ICR mice were given low and high dose (0.015,0.15mg/10g) estradiol pentodate for 7d and dexamethasone 0.15mg for immunosuppression and then inoculated with candida albican to establish a vaginitis model. Colony count of vaginal lavage solution and vaginal histopathological observation were conducted. Results Pentanoic acid estradiol is low, the high dose group of vaginal lavage colony count peaked in the 4th day, respectively 3.324±0.150, 6.825±1.247, and compare the difference was statistically significant (P<0.05), after the injection of dexamethasone pentanoic acid estradiol is low, the high dose group of vaginal lavage colony count in the 7th day peak respectively 8.472±0.954, 14.152±3.24, difference was statistically significant (P<0.01). HE staining showed that there were more PMNs in the estradiol valerate + dexamethasone group. Conclusion Dexamethasone plus estradiol valerate at high dose is the best condition for ICR mouse model of candida albicans vaginitis. And at its best during the week.
