摘 要:目的观察化浊解毒方对溃疡性结肠炎(UC)模型大鼠结肠黏膜病理形态及血清肿瘤坏死因子α(TNF-α)、γ干扰素(IFN-γ)含量的影响,探讨其作用机制。方法将60只雄性Wistar大鼠按照随机数字表法分为正常组15只及造模组45只,造模组采用2,4,6-三硝基苯磺酸(TNBS)/乙醇法诱导UC大鼠模型,并将造模成功的大鼠(42只)按照随机数字表法分为模型组、美沙拉嗪组及化浊解毒方组,每组14只。造模后,模型组予蒸馏水4mL/(kg·d)灌胃;美沙拉嗪组予美沙拉嗪混悬液0.3g/(kg·d)灌胃;化浊解毒方组予化浊解毒方混悬液20g/(kg·d)灌胃。灌胃14d后,比较各组大鼠疾病活动指数(DAI)评分、结肠黏膜损伤指数(CMDI)评分及血清TNF-α、IFN-γ含量。结果与正常组比较,模型组、美沙拉嗪组、化浊解毒方组DAI评分均升高(P<0.05);与模型组比较,美沙拉嗪组、化浊解毒方组DAI评分均降低(P<0.05);与美沙拉嗪组比较,化浊解毒方组DAI评分降低(P<0.05)。与正常组比较,模型组、美沙拉嗪组、化浊解毒方组溃疡、炎症及病变深度评分均升高(P<0.05);与模型组比较,化浊解毒方组溃疡、炎症及病变深度评分均降低(P<0.05),美沙拉嗪组溃疡、炎症评分均下降(P<0.05),病变深度评分无明显变化(P>0.05);与美沙拉嗪组比较,化浊解毒方组溃疡、炎症及病变深度评分均降低(P<0.05)。与正常组比较,模型组血清TNF-α、IFN-γ均升高(P<0.05),美沙拉嗪组、化浊解毒方组血清TNF-α均升高(P<0.05);与模型组比较,美沙拉嗪组、化浊解毒方组血清TNF-α、IFN-γ均降低(P<0.05);与美沙拉嗪组比较,化浊解毒方组血清TNF-α、IFN-γ均降低(P<0.05)。结论化浊解毒方能够调整血清细胞因子,进而调控免疫机制,降低DAI,改善UC模型大鼠结肠黏膜的病理损伤。Objective To observe the effect of Huazhuo Jiedu recipe on colonic mucosa pathology and the serum levels of tumor necrosis factor α(TNF-α) and interferon-γ(IFN-γ) in rats with ulcerative colitis(UC), and to explore its mechanism. Methods According to the random number table method, 60 male Wistar rats were divided into normal group(15 rats) and module group(45 rats). The UC rat model was induced by 2, 4, 6-trinitrobenzenesulfonic acid(TNBS)/ethanol in the module group. 42 successful rats were divided into model group, mesalazine group and Huazhuo Jiedu recipe group according to random number table method, with 14 rats in each group. After modeling, the model group was given 4 mL/(kg·d) distilled water by gavage;the mesalazine group was given 0.3 g/(kg·d) mesalazine suspension by gavage;the Huazhuo Jiedu recipe group was given 20 g/(kg·d) Huazhuo Jiedu recipe suspension by gavage. After 14 days of gavage, the disease activity index(DAI) score, colonic mucosal injury index(CMDI) score, serum TNF-α and IFN-γ contents of rats in each group were compared. Results Compared with normal group, the DAI scores of model group, mesalazine group and Huazhuo Jiedu recipe group were all increased(P<0.05);compared with model group, the DAI scores of mesalazine group and Huazhuo Jiedu recipe group were all decreased(P<0.05);compared with mesalazine group, the DAI score of Huazhuo Jiedu recipe group was decreased(P<0.05). Compared with the normal group, the ulceration, inflammation and lesion depth scores of the model group, mesalazine group and Huazhuo Jiedu recipe group were all increased(P<0.05);compared with model group, the ulceration, inflammation and lesion depth scores in Huazhuo Jiedu recipe group were all decreased(P<0.05), while the ulceration and inflammation of mesalazine group were decreased(P<0.05), with no significant change in lesion depth scores(P>0.05);compared with the mesalazine group, the ulceration, inflammation and lesion depth scores in the Huazhuo Jiedu recipe group were all decreased(P<0.05).
