浸润性乳腺癌组织中RNA结合基序蛋白3的表达水平及意义

摘　　要：目的探究RNA结合基序蛋白3(RBM3)在浸润性乳腺癌患者乳腺癌组织中的表达情况及临床意义。方法收集136例浸润性乳腺癌患者的肿瘤组织和对应癌旁组织。采用实时荧光定量聚合酶链反应法检测乳腺癌组织和癌旁组织中RBM3mRNA的表达情况。根据浸润性乳腺癌患者乳腺癌组织中RBM3mRNA相对表达量的平均值将患者分为RBM3mRNA高表达组(n=61)和RBM3mRNA低表达组(n=75),分析RBM3mRNA表达与浸润性乳腺癌患者临床特征及预后的关系。结果浸润性乳腺癌组织中RBM3mRNA的相对表达量明显高于癌旁组织(P﹤0.01);RBM3mRNA高表达组乳腺癌患者的TNM分期、组织学分级、淋巴结转移数目、雌激素受体(ER)表达情况、孕激素受体(PR)表达情况、人表皮生长因子受体2(HER2)表达情况与RBM3mRNA低表达组患者比较,差异均有统计学意义(P﹤0.05);RBM3mRNA高表达组患者的无瘤生存情况优于RBM3mRNA低表达组(P﹤0.05);RBM3mRNA诊断浸润性乳腺癌患者无瘤生存预后的AUC为0.822(95%CI:0.747~0.882);Cox多因素分析结果显示:TNM分期、肿瘤直径、淋巴结转移和RBM3mRNA表达情况是浸润性乳腺癌患者无瘤生存时间的独立影响因素(P﹤0.05)。结论RBM3mRNA高表达提示浸润性乳腺癌患者预后较好。Objective To investigate the expression and clinical significance of RNA binding motif protein 3 (RBM3) in breast cancer tissues of patients with invasive breast cancer. Method Tumor tissues and adjacent tissues of 136 patients with invasive breast cancer were collected. Real- time fluorescence quantitative polymerase chain reaction was used to detect the expression of RBM3 mRNA in breast cancer tissues and adjacent tissues. According to the mean value of relative RBM3 mRNA expression in breast cancer tissues of patients with invasive breast cancer, patients were divided into the group with high RBM3 mRNA expression (n=61) and the group with low RBM3 mRNA expression (n=75). The relationship between RBM3 mRNA expression and clinical characteristics and prognosis of patients with invasive breast cancer was analyzed. Result The relative expression of RBM3 mRNA in invasive breast cancer tissues and was significantly higher than that in paracancerous tissues (P<0.01). TNM stage, histological grade, number of lymph node metastasis, estrogen receptor (ER) expression, progesterone receptor (PR) expression, and human epidermal growth factor receptor 2 (HER2) expression in patients with high RBM3 mRNA expression group were statistically significant compared with patients with low RBM3 mRNA expression group (P<0.05). The tumor-free survival of invasive breast cancer patients with high expression of RBM3 mRNA was better than that of patients with low expression of RBM3 mRNA (P< 0.05). The AUC of RBM3 mRNA in the diagnosis of tumor- free survival in invasive breast cancer patients was 0.822 (95%CI: 0.747-0.882). Cox multivariate analysis showed that TNM stage, tumor diameter, lymph node metastasis and RBM3 mRNA expression were independent influencing factors for tumor- free survival time of patients with invasive breast cancer (P<0.05). Conclusion The high expression of RBM3 mRNA suggests the better prognosis of patients with invasive breast cancer.
