摘  要:目的:建立耐受顺铂的EB病毒阳性人鼻咽癌细胞系和EB病毒阴性细胞系,并观察顺铂及三氧化二砷对上述细胞增殖的影响。方法:采用小剂量顺铂浓度递增法培养EB病毒阳性的人鼻咽癌细胞系C666-1和EB病毒阴性的人鼻咽癌细胞系CNE-2,基于cck-8法检测三氧化二砷或顺铂对上述亲本细胞及其耐受DDP的细胞系C666-1/DDP、CNE2/DDP的半数抑制浓度(IC50)和对细胞增殖的抑制作用。结果:成功建立耐受顺铂的EBV阳性细胞系C666-1/DDP及EBV阴性鼻咽癌细胞系CNE2/DDP,与亲本细胞在形态上比较无明显差异;顺铂或三氧化二砷对C666-1/DDP细胞的IC50大于对CNE2/DDP细胞(均为P<0.01),且此两种耐药细胞对三氧化二砷的耐药指数差异无统计学意义(P>0.05),这些亲本细胞及其耐药细胞均对三氧化二砷敏感。结论:成功构建耐受顺铂的EBV阳性鼻咽癌细胞系C666-1/DDP和EBV阴性鼻咽癌细胞系CNE2/DDP,均敏感于三氧化二砷。Objective: To establish EBV-positive and negative human nasopharyngeal carcinoma cell lines which are resistant to cisplatin, and observe the effects of cisplatin and arsenic trioxide on the proliferation of cells. Methods: The cisplatin-resistant human nasopharyngeal carcinoma cell lines C666-1 and CNE-2 of EBV were cultured by using gradually increased concentration of small dose cisplatin in culture. The inhibition effects of arsenic trioxide or cisplatin on the above-mentioned parent cells and the IC50 of C666-1/DDP and CNE2/DDP were assessed by CCK8 assays. Results: Compared with the parental cells, there ware no obvious morphological change of the successful establishment of cisplatin-resistant human EBV-positive and negative nasopharyngeal carcinoma cell lines. It showed that the cisplatin or arsenic trioxide were more tolerant to IC50 of C666-1/DDP cells than to CNE2/DDP cells(P<0.01), and there was no obvious difference for the two resistant cell line to the resistance index of arsenic trioxide(P>0.05). Moreover, these parental cells as well as their drug-resistant cells were all sensitive to arsenic trioxide. Conclusion: The successful constructed cisplatin-resistant nasopharyngeal carcinoma cell lines C666-1/DDP and CNE2/DDP are sensitive to arsenic trioxide.
