摘  要:神经鞘氨醇激酶1(SphK1)在激素、生长因子及细胞因子的作用下活化,活化的SphK1促进具有生物活性的脂质信号分子1-磷酸神经鞘氨醇(S1P)的产生,通过激活一种或几种特定的1-磷酸神经鞘氨醇受体(S1PR),从而参与多种生物学行为。SphK1、S1P以及S1PRs在肝癌、胰腺癌、食管癌、胃癌、结直肠癌、乳腺癌、卵巢癌、前列腺癌及白血病等中异常表达,并在肿瘤的发生、发展过程中起重要作用。笔者就SphK1-S1P-S1PRs信号通路在肝癌中的作用和机制以及相关研究进展进行综述。Sphingosine kinase 1(SphK1)activation induced by various hormones, growth factors and cytokines promotes the production of the bioactive lipid signaling molecule sphingosine-1-phosphate(S1 P), which is involved in a variety of biological processes by the activation of one or several sphingosine-1-phosphate receptors(S1 PRs). It has been reported that SphK1, S1 P and S1 PRs are abnormally expressed in some cancers such as hepatocellular carcinoma, pancreatic carcinoma, esophageal cancer, gastric cancer, colorectal cancer, mammary cancer, ovarian cancer, prostatic cancer, leukemia, and the like, playing extremely important roles in the occurrence and development of tumors. The author reviews the roles and mechanisms of SphK1-S1 P-S1 PRs signaling pathway in hepatocellular carcinoma and its related research progress.
