摘　　要：目的探讨喉鳞状细胞癌(LSCC)组织中沉默信息调节因子6(Sirtuin6,SIRT6)的表达水平及临床意义。方法选取2009年1月至2012年1月就诊于我院并接受手术的LSCC患者共50例,病变组织作为实验组,选取16例声带息肉患者的息肉组织作为对照组。应用免疫组织化学染色检测LSCC组织与息肉组织中SIRT6的表达水平,并分析SIRT6的表达与各临床特征之间的相关性。结果免疫组化发现SIRT6在LSCC组织以及息肉组织中均可表达,在LSCC中的表达水平低于对照组(t=-6.326,P<0.001)。SIRT6的表达与分化程度有显著相关性,随着分化程度降低,SIRT6的表达逐渐下降,并存在统计学差异(χ^2=20.576,P<0.001)。SIRT6的表达与临床分期相关,Ⅲ/Ⅳ期SIRT6的表达更低(χ^2=13.675,P<0.05)。SIRT6的表达与有无淋巴结转移无关(χ^2=0.216,P=0.642)。SIRT6高表达患者,无进展生存率更高(χ^2=3.918,P<0.05)。结论SIRT6的表达很可能与LSCC组织的分化、浸润以及预后有关,提示SIRT6或可作为一种新的分子靶点用于喉癌的治疗及预后评估。Objective To evaluate the expression of SIRT6 in laryngeal squamous cell cancer(LSCC)and its clinical significance.Methods 50 LSCC cases who underwent surgery in our hospital from January 2009 to January 2012 were enrolled as the experimental group,and 16 cases with vocal polyps were selected as the control group.Expression of SIRT6 was determined using immunohistochemistry bentween LSCC tissues and polyps.Results Our results demonstrated that the expression level of SIRT6 was significantly decreased in LSCC tissues compared with vocal polyps using immunohistochemistry(t=-6.326,P<0.001).We found that the level of expression of SIRT6 was significantly correlated with the grade of LSCC,and the level of expression of SIRT6 was decreasing with the grade of LSCC malignancy(χ^2=20.576,P<0.001).Additionally,significant relationship was shown between clinical stage and SIRT6 expression which was lower in Ⅲ/Ⅳ stage(χ^2=13.675,P<0.05).The expression of SIRT6 was not signifiantly associated with lymph node metastasis(χ^2=0.216,P=0.642).Furthermore,the high expression of SIRT6 could lead to a longer overall survival time(χ^2=3.918,P<0.05).Conclusion The current study demonstrats that the expression level of SIRT6 may be associated with the clinical-pathological features and prognosis of LSCC.SIRT6 might be a novel biomarker for LSCC diagnosis and prognosis.
