摘  要:目的探究机械张应力通过TGFβ/Smad信号通路介导成骨细胞增殖分化相关因子的表达变化规律。方法采用成骨细胞体外张应力加载模型,使用流式细胞术检测成骨细胞的增殖活性;并提取成骨细胞的mRNA,利用实时聚合酶链反应(RT-PCR)技术检测TGFβ/Smad及成骨分化相关因子ALP、OCN、Runx2、COL-I的mRNA表达水平。结果机械张应力促进了成骨细胞的增殖活性,并同时改变了TGFβ/Smad、ALP、OCN、Runx2及COL-I mRNA的表达水平。结论 TGFβ/Smad信号通路可能参与机械应力对成骨细胞增殖分化活性的调控。Objective The present research was performed to investigate the modulator expression of TGFβ/Smad signaling pathway-mediated osteoblasts proliferation and differentiation caused by mechanical straining. Methods The tensional stress-stretched osteoblasts model was applied for the in vitro study. The proliferation of osteoblasts was detected using flow cytometry. The mRNA expressions of TGFβ/Smad, ALP, OCN, Runx2 and COL-I were measured by reverse transcription-polymerase chain reaction(RT-PCR). Results The mechanical tensional stress promoted the osteoblasts proliferation and regulated the expressions of TGFβ/Smad, ALP, OCN, Runx2 and COL-I mRNA. Conclusion TGFβ/Smad pathway might be involved in the regulation of mechanical stress-induced osteoblasts proliferation and differentiation.
