摘 要:目的探讨昆布多糖硫酸酯(laminaria sulfate,LAMS)对心肌缺血再灌注模型大鼠心肌损伤的保护作用及其机制。方法采用冠状动脉结扎法建立心肌缺血再灌注(I/R)大鼠模型,适应性饲养合格的大鼠随机分成5组:假手术组(Ctrl组)、模型组(I/R组)、I/R+LAMS(25mg)组、I/R+LAMS(50mg)组和I/R+LAMS(100mg)组。测定平均动脉压(MAP)、左室收缩压(LVSP)和心率(HR);对心肌组织进行HE染色和TUNEL染色观察心肌损伤情况;ELISA检测炎症标记分子、心肌损伤标记物和氧化应激指标含量;蛋白印迹法检测心肌组织中转录因子NF-E2相关因子(Nrf2)、血红素氧合酶-1(HO-1)和醌氧化还原酶1(NQO1)蛋白表达水平。结果昆布多糖硫酸酯能降低心肌缺血再灌注模型大鼠的心肌损伤程度,上调MAP、HR和LVSP水平(P<0.01);降低心肌细胞凋亡率(P<0.01),抑制Caspase-3、Caspase-9高表达(P<0.01);抑制IL-6、IL-1β和iNOS表达上调(P<0.01);抑制CK-MB、cTnⅠ和Mb高表达(P<0.01);抑制Nrf2、HO-1和NQO1表达下调(P<0.01)。结论昆布多糖硫酸酯能通过Nrf2信号通路对心肌缺血再灌注模型大鼠的心肌损伤起到保护作用。Objective To study the protective effect of laminarin sulfate (LAMS) on myocardial injury in ischemia-reperfusion model rats and its mechanism. Methods The model of myocardial ischemia-reperfusion was established by coronary artery ligation in rats. The rats were randomly divided into control group,model group (I/R group),I/R+LAMS group (25 mg),I/R+LAMS group (50 mg) and I/R+LAMS group (100 mg). The mean ventricular systolic pressure (MAP),left ventricular systolic pressure (LVSP) and heart rate (HR) were measured. Myocardial tissue injury was detected by HE staining and TUNEL staining. Myocarditis marker molecule,cardiac injury markers and oxidative stress factor were detected by ELISA. The expression levels of transcription factor NF-E2 related factor (Nrf2),heme oxygenase-1 (HO-1) and quinone oxidoreductase-1 (NQO1) were detected by Western blotting. Results LAMS could reduce the injury of myocardial tissues,increase the levels of MAP,HR and LVSP( P <0.01);reduce the apoptosis rate of myocardial cells ( P <0.01),inhibit the overexpressions of Caspase-3 and Caspase -9 ( P <0.01);inhibit the expressions of IL-6,IL-1 beta and iNOS ( P <0.01),and the overexpression of CK-MB,cTnⅠ and MB ( P <0.01);and up-regulate Nrf2,HO-1 and NQO1 ( P <0.01). Conclusion LAMS can protect the myocardium from ischemia-reperfusion injury in rats through Nrf2 signaling pathway.
