摘 要:目的研究HOXB7基因敲减对裸鼠结肠癌肝转移模型的抑癌机制。方法将转染pcDNA3.1-HOXB7敲减或pcDNA3.1-scramble重组质粒的人结肠癌细胞株HT29经裸鼠脾脏接种建立稳定的肝转移模型。参与模型的实验裸鼠共20只,其中10只HOXB7基因敲减鼠列为研究组,10只scramble鼠列为对照组。制模4周后解剖观察两组裸鼠模型肝转移瘤的组织形态学和组织病理学变化。结果裸鼠结肠癌肝转移模型的成瘤率为100%(20/20)。组织形态学角度分析:与对照组比较,研究组显著抑制裸鼠模型的肝转移瘤形成;递减或衰减肝转移瘤的大小、肝体质量、病灶间融合、肝表面隆凸及肝转移评分,差异有统计学意义(P<0.05)。组织病理学角度分析:与对照组比较,研究组肿瘤细胞排列稀疏、边缘型分布、浸润深度浅、中央区坏死或凋亡成片,差异有统计学意义(P<0.05)。结论HOXB7基因敲减可有效抑制裸鼠模型的结肠癌肝转移瘤形成,至于具体的抑癌模式和量化标准的制定需有待于进一步考证。Objective To study the anti-cancer mechanism of HOXB7 gene knockdown on liver metastasis model of colon cancer in nude mice. Methods HT29 Human colon cancer cells transfected by pcDNA3.1-HOXB7 knockdown of recombinant vector or pcDNA3.1-scramble of vector were injected into spleen of nude mice used to establish a stable hepatic metastasis of colon cancer nude mice model.A total of twenty nude mice were designed into two groups,and ten nude mice of HOXB7 gene knockdown named study group,and ten nude mice of scramble group named control group.Histomorphology and histopathological changes of liver metastases were detected of the two groups after 4 weeks of modeling. Results The tumor formation rate was 100%(20/20) of liver metastasis model of colon cancer in nude mice.For the histomorphological analysis compared with the control group,the study group significantly inhibited the liver metastasis in the nude mice model with a statistical difference( P <0.05).The scoring was significantly distinguished( P <0.05),on the size of the decreased or attenuated liver metastases,liver mass,interfocal fusion,liver surface eminence and liver metastasis.For the histopathological analysis,the liver metastasis of study group was microscopically distinct from the control group( P <0.05),which displayed loosely arranged cells with marginal distribution and less invasion in the edge of the tumor,as well as necrosis and cell apoptosis in the center. Conclusion HOXB7 gene knockdown can effectively inhibit the formation of liver metastases in nude mice with colon cancer.As for the specific tumor suppression model and the development of quantitative criteria,further research is needed.
