摘  要:目的探讨锰苯甲酸卟啉(MnTBAP)在蛛网膜下腔出血(SAH)早期脑损伤中的保护作用。方法将100只SD大鼠随机分为空白对照组、假手术组、模型组(SAH)和实验组(SAH+MnTBAP),每组25只。比较各组大鼠的神经功能缺陷评分、脑组织含水含量、血脑屏障通透性(Evans蓝外渗实验)、线粒体三磷酸腺苷合成酶6亚基(ATPase-6)和细胞色素C蛋白(Cytc)表达及丙二醛(MDA)含量、超氧化物歧化酶(SOD)和谷胱甘肽过氧化物酶(GSH-Px)活性的差异。结果与模型组相比,实验组的神经功能缺陷评分明显增高,脑水含量明显减少,血脑屏障通透性降低,ATPase-6蛋白表达上调,Cytc表达下降,MDA含量减少,SOD和GSH-Px活性增加。结论MnTBAP能够改善神经功能并对线粒体具有保护作用,MnTBAP在SAH早期脑损伤中可能具有神经保护作用。Objective To investigate the protective effect of MnTBAP on early brain injury following subarachnoid hemorrhage in rat model. Methods 100 SD rats were randomly divided into four groups: the blank control group,the sham operation group,the model group (SAH) and the experimental group (SAH+MnTBAP),25 rats per group.The difference among these groups of the neurological deficit score,brain water content,blood-brain barrier permeability (Evans blue extravasation test),ATPase-6 and cytochrome C protein expression,MDA,SOD and GSH-Px active level was observed 24 h after SAH. Results Compared with the model group,the neurological deficit score was significantly increased,brain water content was significantly decreased,blood-brain barrier permeability was significantly decreased,ATPase-6 protein expression was significantly increased,cytochrome C protein expression was significantly decreased,MDA content was significantly decreased,SOD and GSH-Px activity were significantly increased in the experimental group. Conclusion MnTBAP can improve neurological function and protect mitochondria.MnTBAP may play a neuroprotective role in early brain injury following SAH.
