摘 要:目的在一些恶性肿瘤中,Notch和Hedgehog信号通路之间存在着交互对话,本研究目的是探讨沉默Notch1基因对三阴性乳腺癌MDA-MB-231细胞Hedgehog信号通路表达的影响。方法应用siRNA干扰MDA-MB-231细胞Notch1基因后,用RT-PCR检测Notch1、下游靶基因Hes1以及Hedgehog信号通路中Smo、Gli1的mRNA表达情况,应用Western Blot方法检测Notch1、Hes1以及Hedgehog信号通路中Smo、Gli1的蛋白表达情况。结果siRNA沉默Notch1基因后,与空白对照组和阴性对照组相比,实验组Notch1、Hes1、Smo、Gli1的mRNA与蛋白表达水平均有明显降低(P<0.01)。结论在三阴性乳腺癌MDA-MB-231细胞中,沉默Notch信号通路可抑制Hedgehog信号通路的活性,两信号通路之间存在着交互对话。Objective There was cross-talk between Notch and Hedgehog signal pathway in some malignant tumors.The purpose of this study was to investigate the effect of silencing Notch1 gene by small interfering RNA(siRNA)on the expression of the Hedgehog signaling pathway in triple-negative breast cancer MDA-MB-231 cells.Methods MDA-MB-231 cells was interfected by the Notch1 siRNA with lipofectamine transfection.The mRNA expressions of Notch1,Hesl,Smo and Glilwere detected by RT-PCR,the protein expressions of Notch1,Hesl,Smo and Glil were detected by Western Blot.Results Silencing Notch1 of Notch signaling pathways in the MDA-MB-231 cells decreased the mRNA and protein expressions of Notch1,Hesl,Smo and Glil.there was statistically significant difference between experimental group and control groups(P<0.01).Conclusion Silencing Notch1 of the Notch signaling pathways may inhibit Hedgehog signaling pathway,there may be cross-talk between Notch signaling pathway and Hedgehog signaling pathway in triple-negative breast cancer.
