摘 要:目的:研究白花败酱草醇提取液抑制小鼠移植性宫颈癌的作用及机制。方法:对败酱草大、小两个剂量醇提取液抑制U14荷瘤小鼠肿瘤生长的比率以及对胸腺和脾腺指数的影响的观察;采用比色分析法观察血清总抗氧化能力(T-AOC)和超氧化物歧化酶(SOD)活性以及丙二醛(MDA)的变化情况;本研究采用HE染色在光镜下,对败酱草引发并诱导肿瘤组织细胞不同分化程度观察。结果:从高、低剂量败酱草醇提取液均能在一定程度上抑制U14宫颈癌细胞的生长,它们分别的抑瘤率是54.23%、49.19%;同时观察到荷瘤鼠胸腺和脾腺指数发生明显降低,并且败酱草醇提取液还能够有效降低U14荷瘤鼠血清中MDA的水平,并且同时对其提高T-AOC和SOD的活性。经HE处理染色后,其形态学上观察败酱草醇提取液针对肿瘤组织形态能在一定程度上改善达到控制作用。结论:白花败酱草醇提取液有控制以及拮抗并改善U14宫颈癌发展的作用,其发生机制可能与调动并调节小鼠免疫功能,以及提高抗氧化防御系统中各酶的活性有关。Objective:To investigate the anti-tumor effect of the alcohol extract of Patrina Villosa Juss (PVJ) to cervical carcinoma cells in mice U14 and its mechanism. Methods:We used mice sarcoma U14 as research sample and deal with every group with the water extract of PVJ (high and low dosage). We measured the rate of inhibiting tumor and the thymus index, spleen index, to detect the activities of T-AOC, SOD, and MDA in serum with colorimetric method. After He dye, the patrina villosa juss to observe sarcoma cell under the optical microscope, can induce tumor cells disintegration to some extent. Results:PVJ could increase ratios of tumor inhibited in U14-bearing mice. The inhibitory rate of high and low dosage of the alcohol extract of PVJ group is 54.23% and 49.19%. Thymus index and Spleen index could obviously decrease. Compared with the control group, the alcohol extract of PVJ could increase the activities of T-AOC, SOD in serum,and decreased the content of MDA. After HE dye, Pathological diagnosis indicates that the alcohol extract of PVJ can improve the tumor organization modality. Conclusion:the alcohol extract of PVJ may be obviously inhibiting mice sarcoma U14 of cervical cancer. The mechanism of action may be regulating immunity system function, increasing the activity of anti-oxidationase and inhibiting the growth of tumor cells.
