摘 要:为揭示青钱柳内生真菌的分布、多样性及其代谢产物的抑菌活性,并筛选产黄酮的内生真菌,挖掘植物内生真菌新资源,采用组织分离的方法对青钱柳的枝、皮、叶、根部位的内生真菌进行分离,用马丁氏培养基、察氏培养基、PDA培养基对内生真菌进行培养,通过观察内生真菌的形态特征进行初步分类,同时提取其发酵产物,并以金黄色葡萄球菌、大肠杆菌、白色念珠菌为供试病原菌对分离的内生真菌进行抑菌活性试验,通过显色试验和薄层层析试验筛选出产黄酮的内生真菌.试验共分离得到67株内生真菌,通过菌落形态和显微形态对分离得到的青钱柳内生真菌进行分类,其分别隶属于3纲4目6科10属;青钱柳不同组织中内生真菌的分离结果不同,其中根部分布的内生真菌从数量和种属上都占优势;马丁氏培养基培养的菌株种类少、数量多,而察氏培养基和PDA培养基培养的菌株种类较多、菌株较少;抑菌试验共得到13株具有抑菌作用的内生真菌,其中有2株内生真菌的发酵产物对供试菌株均表现出较强的抑菌活性,分别属于青霉属和链格孢属;并且筛选出7株菌株(PP03、PY01、PG11、PZ06、PY12、PP12和PP06)具有稳定的产生黄酮类化合物的能力.In order to clarify the distribution and diversity of endophytic fungi from Cyclocaryapaliurus (Batal) Ijinskaja, and explore its potential application as source of flavonoid, endophytic fungi in different parts of Cyclocaryapaliurus were isolated by tissue separation method, and morphological identification was conducted by culturing them in Martin′s medium, Catch′s medium and PDA medium. Staphyloccocus aureus Rosenbach, Escherichia coli, Bacillus subtilis, Moniliaalbican were used as pathogenic microbe to evaluate the antibacterial activity, and flavonoid-producing endophytic fungi was screened by color test and thin layer chromatography. The results showed that 67 strains of endophytic fungi were obtained and belonged to 10 genera, 6 families, 4 orders and 3 classes. Distribution and abundance of endophytic fungi varied significantly among different tissues, with the largest quantity and highest diversity coming from the root. Strains cultured in Martin′s medium had fewer types but larger quantity, while Catch′s medium and PDA culture medium resulted in less strains but more types. A total of 13 strains with antifungal activity were screened. Among them, the metabolites of 2 strains, which belonged to Trichoderma and Penicillium, respectively, showed notable antibacterial activity against the tested microbes. Seven strains (PP03, PY01, PG11, PZ06, PY12, PP12 and PP06) were screened for their able ability to produce flavonoids.
