星点设计响应面法优化纳米结构脂质载体载辛伐他汀的研究

摘  要:目的利用星点设计响应面法优化制备纳米结构脂质载体载辛伐他汀的最佳制备条件。方法 采用星点设计响应面法,以乳剂的平均粒径、多分散指数和zeta电位为评价指标,考察处方中自变量OA浓度、P188浓度和微射流次数对制剂的影响,预测出最优制备条件。结果 优选出的最佳制备条件:OA浓度为40%(wt%),P188浓度为0.4%(w/v%),微射流机循环次数为4次。依据最佳制备条件完成纳米结构脂质载体载辛伐他汀的制备,平均粒径为174.7 nm,包封率94.1%,实际载药量18.2。结论 星点设计响应面分析法所建立的模型能较好地用于纳米结构脂质载体载辛伐他汀的处方优化,纳米结构脂质载体可以很好地实现SV包裹。Objective To optimize the preparation conditions of the nanostructured lipid carrier containing simvastatin by the star point design response surface method. Methods The effects of the OA concentration, P188 concentration and the number of microjet on the preparation were examined with the average particle size, polydispersity index and zeta potential of the emulsion as the evaluation index using the star point design response surface method. Then the optimal preparation conditions were predicted. Results The best preparation conditions were OA concentration of 40%(wt%), P188 surfactant concentration of 0.4%(w/v%), and the frequency of microjet cycle of 4 times. Simvastatin loaded on nanostructured lipid carrier was prepared according to the optimum preparation conditions. The average particle size was 174.7 nm, the encapsulation efficiency was 94.1%, and the actual drug loading was 18.2%. Conclusion The model established by the star point design response surface analysis method can be used to optimize the formulation of the nanostructured lipid carrier loaded on nanostructured lipid carriers, and the nanostructured lipid carriers can effectively encapsulate SV.
