摘　　要：目的探讨PDK-1蛋白在胃癌组织中的表达及其临床意义。方法选择胃癌患者80例作为研究对象,采用酶联免疫吸附法及免疫组化法测定胃癌组织及癌旁黏膜组织中PDK-1蛋白的表达水平,分析其临床意义。结果胃癌组PDK-1蛋白表达水平高于癌旁组(t=68. 733,P <0. 01);胃癌组PDK-1阳性率高于癌旁组(χ^2=81. 667,P <0. 01)。PDK-1蛋白表达与胃癌患者年龄相关性不明显(χ^2=0. 752,P> 0. 05);与病理分型、肿瘤最大径、病理学分级、TMN分期、浸润深度、淋巴血管间隙浸润、淋巴结转移、复发相关性明显,且鳞癌、肿瘤最大径≥5 cm、病理学分期越高、TMN分期越高、浸润深度越深、有淋巴血管间隙浸润、有淋巴结转移、有复发,PDK-1蛋白阳性表达率越高(χ^2=34. 504、4. 969、11. 709、15. 038、8. 130、6. 771、7. 326、8. 252,P <0. 05或P <0. 01)。PDK-1阴性组3年生存率及生存期均明显高于PDK-1阳性组(χ^2=8. 604、Z=15. 908,P <0. 05或P <0. 01)。结论 PDK-1蛋白在胃癌组织中表达量增加,PDK-1在胃癌的发生发展过程中起促进作用;PDK-1蛋白低表达的胃癌患者能获得较好的预后。Objective To investigate expression of PDK-1 protein in gastric carcinoma and its clinical significance.Methods The gastric cancer tissues and adjacent mucosal tissues of 80 patients with gastric cancer were selected,then the expression level of PDK-1 protein and its clinical significance were analyzed. Results The expression of PDK-1 protein in gastric cancer group was higher than that in paracancer group( t = 68. 733,P < 0. 01);The positive rate of PDK-1 in gastric cancer group was higher than that of paracancer group(χ^2= 81. 667,P < 0. 01);The PDK-1 protein expression was not correlated with the age(χ^2= 0. 752,P > 0. 05),but was positively correlated with the pathological type,maximum diameter of tumor(≥5 cm),pathological grade,TMN stage,depth of invasion,infiltration of lymphatic vessels,lymph node metastasis,and recurrence(χ^2= 34. 504,4. 969,11. 709,15. 038,8. 130,6. 771,7. 326,8. 252,P < 0. 05,P < 0. 01). Conclusion PDK-1 protein is highly expressed in gastric cancer tissue,which plays a catalytic role in the development of gastric cancer,so the low expression of PDK-1 protein in gastric cancer patients can get a better prognosis.
