胃癌组织中长链非编码RNA ZDHHC8P1的水平及临床意义

摘　　要：目的 探讨胃癌组织中长链非编码RNA(lncRNA)DHHC型锌指蛋白8假基因1(ZDHHC8P1)的表达及其临床意义。方法 收集2013年1月至2017年12月我院收治的128例胃癌患者的胃癌组织及对应癌旁组织标本,应用实时定量PCR(QPCR)检测胃癌组织及癌旁组织中ZDHHC8P1的表达,分析其与临床病理特征的关系。根据随访资料分析ZDHHC8P1表达与胃癌预后的关系。结果 QPCR检测结果显示,胃癌组织中lncRNAZDHHC8P1相对表达量为6.114±1.849,高于癌旁组织的1.532±0.638(P<0.05)。胃癌组织中ZDHHC8P1相对表达量与性别、年龄和肿瘤位置无关,而与肿瘤直径、分化程度、浸润深度、淋巴结转移、远处转移和TNM分期有关。单因素分析发现ZDHHC8P1低表达患者的中位总生存期(OS)为52.0(95%CI:44.059~59.941)个月,长于高表达者的35.0(95%CI:36.549~45.451)个月(P<0.05);经Cox多因素回归分析发现ZDHHC8P1表达是影响胃癌OS的独立因素(OR=2.321,95%CI:1.332~4.045,P=0.003)。结论 ZDHHC8P1在胃癌组织中表达上调,其表达水平升高与胃癌的恶性程度和预后有关,在胃癌诊治中有一定应用前景。Objective To investigate the expression of long-chain non-coding RNA zinc finger DHHC-type containing 8 pseudogene 1 (ZDHHC8P1) in gastric cancer and its clinical significance.Methods One hundred and twenty-eight cases of gastric cancer tissues and corresponding paracancerous tissues from January 2013 to December 2017 were collected in our hospital.Real-time quantitative PCR(QPCR) was used to detect the expression of ZDHHC8P1 in gastric cancer tissues and adjacent tissues,and the correlation between ZDHHC8P1 and clinicopathological characteristics was analyzed.The relationship between ZDHHC8P1 expression and prognosis of gastric cancer was analyzed by Cox risk ratio model based on follow-up data.Results The results of QPCR showed that the relative expression of ZDHHC8P1 in gastric cancer tissues was 6.114±1.849,higher than 1.532±0.638 in adjacent tissues (P<0.05).The relative expression of ZDHHC8P1 in gastric cancer tissues was not related to sex,age and location of tumors,but related to tumor diameter,differentiation degree,depth of invasion,lymph node metastasis,distant metastasis and TNM stage.Univariate analysis showed that the median overall survival of ZDHHC8P1 was 52.0 (95% CI: 44.059-59.941) months,which was longer than 35.0 (95% CI: 36.549-45.451) months in the high expression group (P<0.05).The expression of ZDHHC8P1 was found to be an independent prognostic factor for gastric cancer (OR=2.321,95%CI: 1.332-4.045,P= 0.003).Conclusion The expression of ZDHHC8P1 was up-regulated in gastric cancer tissues,which was related to the malignant degree and prognosis of gastric cancer.It has certain application prospects in the diagnosis and treatment of gastric cancer.
