肝动脉灌注P38MAPK抑制剂对大鼠肝囊性包虫病的治疗作用

摘  要:目的探讨肝动脉灌注P38MAPK抑制剂对大鼠肝囊性包虫病的治疗作用。方法选取成功接种细粒棘球蚴于肝脏实质内体重相近的清洁级雌性Wistar大鼠60只(标本为80只),随机分为生理盐水对照组(A组),50μmol/LSB-202190组(B组),100μmol/LSB-202190组(C组),每组20只。A、B、C组分别经肝动脉灌注0.3mL生理盐水、50μmol/LSB-202190溶于0.3mL生理盐水、100μmol/LSB-202190溶于0.3mL生理盐水。分别于灌药结束后1、3、7、14、42d采集大鼠外周静脉血检测谷丙转氨酶(ALT)、谷草转氨酶(AST)、葡萄糖(Glu)、三酰甘油(TG)、总蛋白(TP)、清蛋白/球蛋白比值(A/G)、清蛋白(Alb)、总胆红素(TBIL)等肝功能指标。42d处死全部大鼠,取细粒棘球蚴感染成功的肝组织,固定于10%甲醛溶液中。经包埋连续切片每个标本5张脱蜡、染色后观察病理变化。结果B、C组大鼠ALT、AST与A组比较,差异均有统计学意义(P<0.05);B组大鼠ALT,AST与C组比较,差异有统计学意义(P<0.05);A组大鼠Glu与B、C组比较,差异均有统计学意义(P<0.05);、B组大鼠TG与A、C组比较,差异均有统计学意义(P<0.05);B组大鼠A/G与C组比较,差异有统计学意义(P<0.05);C组大鼠Alb与A、B组比较,差异均有统计学意义(P<0.05);A组大鼠TBIL与B、C组比较,差异均有统计学意义(P<0.05);B组大鼠TBIL与C组比较,差异有统计学意义(P<0.05)。提示肝动脉灌注P38MAPK抑制剂SB-202190后对感染囊性包虫大鼠的肝功能具有明显改善作用。与A组比较,SB-202190组大鼠病理切片观察到肝细胞纤维化较轻,炎性细胞浸润较少,可见细胞轮廓。结论SB-202190试剂对大鼠肝包虫病治疗具有明显的改善作用。Objective To investigate the therapeutic effect of hepatic artery infusion of P38MAPK inhibitor on hepatic cystic echinococcosis in rats. Methods Sixty clean female Wistar rats(80 specimens) were randomly selected and inoculated with Echinococcus granulosus in the liver parenchyma.The rats were randomly divided into three groups:saline control group A,50 micromol/L SB-202190 group B,100 micromol/L SB-202190 group C,A,B and C groups infused 0.3 mL saline via hepatic artery,50 micromol/L SB-202190 was dissolved in 0.3 mL saline,100 micromol/L SB-202190 was dissolved in 100 micromol saline,respectively.L/L SB-202190 was dissolved in 0.3 mL saline.Peripheral venous blood of rats was collected at 1,3,7,14,42 d after the end of administration to detect the liver function indexes such as alanine aminotransferase(ALT),aspartate aminotransferase(AST) and total bilirubin(TBIL).All rats were sacrificed for 42 days.The liver tissues infected with Echinococcus granulosus were taken and fixed in 10% formaldehyde solution.Pathological changes were observed after 5 pieces of each specimen were dewaxed and stained. Results The results of ALT and AST in group B and C were significantly different from those in group A, P <0.01 Significance;There was significant difference between group B and C in the result of A/G P <0.01;group C and group A and B in the result of Alb, P <0.05;group A,B and group A and C in the result of TBIL, P <0.05,These results suggest that hepatic artery infusion of P38MAPK inhibitor SB-202190 can significantly improve the liver function of rats infected with cystic echinococcosis.Compared with saline group,pathological sections of SB-202190 group showed less fibrosis,less inflammatory cell infiltration and cell contour.SB-202190 group can significantly improve liver function.Compared with saline group,pathological sections of SB-202190 group showed less fibrosis,less inflammatory cell infiltration and cell contour.
