摘  要:目的 探讨肝癌缺失基因(DLC-1)和埃兹蛋白(Ezrin)在结肠癌中的表达及其与结肠癌根治术后复发的关系。方法 选取72例结肠癌患者为研究对象,所有患者均行结肠癌根治术。取术中切除的结肠癌组织及癌旁正常组织,qRT-PCR法检测结肠癌及癌旁组织中DLC-1与Ezrin的mRNA相对表达量;免疫组化法检测Ezrin蛋白表达水平;分析DLC-1、Ezrin表达水平与结肠癌患者临床病理参数的关系。采用Cox比例风险模型对DLC-1、Ezrin表达与结肠癌术后复发的相关因素进行关联分析。结果 与癌旁组织相比,结肠癌组织中DLC-1mRNA表达水平及蛋白阳性表达率均显著降低(P<0.05),而EzrinmRNA表达水平及蛋白阳性表达率均显著升高(P<0.05);DLC-1、Ezrin表达均与结肠癌血管浸润、神经侵犯、淋巴结转移、分化程度、TNM分期明显相关(χ^2=10.307,P<0.05);DLC-1阴性表达及Ezrin阳性表达患者术后复发率均显著升高(P<0.05);Cox分析显示淋巴结转移、DLC-1蛋白表达、Ezrin蛋白表达均为影响结肠癌术后复发的危险因素。结论 结肠癌中DLC-1表达下调,而Ezrin表达上调,且均可增加术后复发的风险。Objective To investigate the expressions of deleted in liver cancer-1(DLC-1) and Ezrin in colon cancer and their relationships with recurrence after radical resection. Methods Seventy-two patients with colon cancer were selected as the study subjects, the colon cancer tissues and adjacent tissues resected by radical resection were taken as the colon cancer group and the control group respectively. QRT-PCR method was used to detect the relative expressions of DLC-1 and Ezrin mRNAs in colon cancer and adjacent tissues;the expressions of DLC-1 and Ezrin proteins were detected by immunohistochemistry;the clinicopathological parameters of colon cancer patients were collected, the relationships between DLC-1 and Ezrin expressions with clinicopathological parameters wrtr analyzed. Cox proportional hazards model was used to analyze the factors influencing recurrence after radical resection of colon cancer. Results Compared with the control group, the expression level of DLC-1 mRNA and the positive protein expression rate in colon cancer group were significantly lower (P<0.05), the expression level of Ezrin mRNA and the positive protein expression rate were significantly increased (P<0.05);the expressions of DLC-1 and Ezrin were significantly correlated with vascular invasion, nerve invasion, lymph node metastasis, differentiation and TNM stage (P<0.05);the recurrence rate of patients with negative DLC-1 expression was significantly increased (P<0.05), the recurrence rate of Ezrin positive patients increased significantly (P<0.05);Cox analysis showed that the expression of DLC-1Ezrin protein was a risk factor for postoperative recurrence of colon cancer. Conclusion Low expression of DLC-1 and high expression of Ezrin in colon cancer can increase the risk of recurrence after radical resection, and can participate in the occurrence and development of colon cancer.
