摘 要:目的分析放射敏感性不同食管癌细胞株长链非编码RNA HOTAIR的表达水平及其与放射敏感性之间的关系。方法以3种食管癌细胞(KYSE510、ECA109、KYSE410细胞)为研究对象。采用RT-qPCR检测细胞中HOTAIR mRNA表达水平,采用平板克隆形成实验检测不同剂量X射线照射下的存活分数(survival fraction, SF)。采用 t 检验或方差分析差异,Pearson法进行相关性分析。结果 3种食管癌细胞(KYSE510、ECA109、KYSE410)2 Gy照射细胞SF2分别为0.38±0.04、0.63±0.03、0.67±0.05,KYSE410的放射敏感性最低,KYSE510放射敏感性最高。HOTAIR mRNA表达水平与SF2呈正相关( r =0.790, P <0.05)。KYSE510细胞HOTAIR表达水平放疗前比放疗后高2.30倍,KYSE410细胞HOTAIR表达水平放疗后比放疗前高2.81倍。结论 HOTAIR可能成为食管癌细胞放疗敏感性的预测分子,其高表达提示放疗抵抗。Purpose To investigate the expression of long non-coding RNA HOTAIR in esophageal cancer cell lines with different radiosensitivity and the relationship between the expression level of HOTAIR and cellular radiosensitivity. Methods Three esophageal cancer cell lines (KYSE510, ECA109, KYSE410) were used in this study. Quantitative real-time polymerase chain reaction was applied to measure the expression of HOTAIR in the above cell lines, colony forming assay was used to measure the survival fraction (SF) of different cells irradiated by different doses of X-ray. The t-test or analysis of variance was applied for analysis of differences. The correlation analysis was applied by Pearson methods. Results The SF2 of KYSE510, ECA109 and KYSE410 was 0.38±0.05, 0.63±0.04 and 0.67±0.06, respectively. The radiosensitivity of KYSE410 was the lowest, and the radiosensitivity of KYSE510 was the highest. The expression level of HOTAIR was positively correlated with SF2 (r=0.879, P<0.05). The expression level of HOTAIR in KYSE510 before radiotherapy were 2.30 times higher than the expression after radiotherapy. The expression level of HOTAIR in KYSE410 after radiotherapy were 2.81 times higher than the expression before radiotherapy. Conclusion HOTAIR may be used as the predictive marker of radiosensitivity of esophageal cancer cells, and its higher expression suggests radiation resistance.
