摘 要:[目的]筛选并优化能够降解农村生活污水中有机物的高效降解菌。[方法]利用普遍适用性培养基和选择性培养基进行菌株的分离筛选,以化学需氧量为评判指标进行复筛、鉴定及高效复合菌的初步构建,通过单因素及正交试验优化高效降解菌的发酵条件。[结果]从35株初筛菌株中复筛出5株有机物降解率在40%以上的菌株,最优发酵条件为:接种量0.3%、发酵时间5d、复合菌的配比B2(假单胞菌)∶N1(棒状杆菌)为4∶1时,有机物降解率可达84.46%。[结论]该研究可为农村生活污水的治理提供参考。[Objective] To screen and optimize highly efficient bacteria which can degrade organic matter in rural domestic sewage.[Method] The strains were isolated and screened by universal applicability medium and selective medium. The chemical oxygen demand (COD) was used as the evaluation index for rescreening, identification and preliminary construction of high-efficiency composite bacteria. The fermentation conditions of high-efficiency degradation bacteria were optimized by single factor and orthogonal test.[Result] Among the 35 strains, 5 strains with more than 40% degradation rate of organic matter were screened out. The optimum fermentation conditions were inoculation amount of 0.3%, fermentation time of 5 days, and the ratio of B 2 ( Pseudomonas sp.) to N 1 ( Corynebacterium sp.) was 4 ∶1, and the degradation rate of organic matter could reach 84.46%.[Conclusion] The study can provide reference for the treatment of rural domestic sewage.
