摘 要:目的探讨荔枝核总黄酮(TFL)对大鼠结直肠癌前病变的影响。方法将40只大鼠随机分为对照组、模型组、TFL小剂量组、TFL大剂量组。其他组大鼠给予皮下注射二甲肼20mg/kg,对照组大鼠只注射等剂量生理盐水。注射二甲肼次日起,对照组、模型组给予生理盐水灌胃,TFL小剂量组、TFL大剂量组分别按100mg/(kg·d)、200mg/(kg·d)给予TFL灌胃。造模第15周后,取大鼠全部结直肠在镜下观察异性隐窝灶(ACF)的数量,并检测ACF处黏膜程序性死亡受体1(PD-1)、核因子-кB(NF-кB)p65mRNA的表达水平,同时检测血清谷胱甘肽的含量。结果对照组未发现有ACF病变,模型组、TFL小剂量组、TFL大剂量组ACF的数量依次下降(均P<0.05)。模型组、TFL小剂量组、TFL大剂量组、对照组大鼠血清谷胱甘肽含量依次升高(均P<0.05)。模型组ACF中PD-1mRNA的表达水平高于对照组及TFL大剂量组(P<0.05),其余组间差异均无统计学意义(均P>0.05)。与对照组比较,模型组NF-κBp65mRNA的表达水平升高,而TFL大剂量组的表达水平下降(均P<0.05);模型组、TFL小剂量组、TFL大剂量组NF-κBp65mRNA的表达水平依次降低(均P<0.05)。结论TFL对二甲肼诱导的大鼠结直肠癌前病变具有良好的抑制作用,大剂量TFL的作用更佳。NF-κBp65可能为TFL抑制ACF形成的关键因子。Objective To explore the effect of total flavone of semen litchi(TFL)on colorectal precancerous lesion in rats.Methods Forty rats were randomly divided into control group,model group,low-dose TFL group and high-dose TFL group.Hypodermic injection of 1,2-dimethylhydrazine(20 mg/kg)was conducted in the rats of the other groups,and injection of normal saline with equal volume in the rats of the control group.From the second day after administration of 1,2-dimethylhydrazine,the control group and the model group received normal saline by gavage,the low-and high-dose TFL groups were treated by irrigation of TFL with 100 mg/(kg·d)and 200 mg/(kg·d),respectively.At the 15th week after modeling,the entire colorectums of the rats were removed to observe aberrant crypt foci(ACF)under microscope,and the expression levels of programmed cell death protein 1(PD-1)and nuclear factor-kappaB(NF-κB)p65 mRNAs in the ACF mucosae were detected,and the content of serum glutathione was also measured.Results No ACF lesion was found in the control group,and the amount of ACF decreased in the order of the model group,the low-dose TFL group and the high-dose TFL group(all P<0.05).The content of serum glutathione increased in the order of the model group,the low-dose TFL group,the high-dose TFL group and the control group(all P<0.05).The model group had a higher expression level of PD-1 mRNA in the ACF compared with the control group and the high-dose TFL group(P<0.05),no statistically significant difference was observed among the other groups(all P>0.05).The expression level of NF-κBp65 mRNA increased in the model group,but decreased in the high-dose TFL group compared with the level in the control group(all P<0.05);the expression level of NF-κBp65 mRNA decreased in the order of the model group,the low-dose TFL group and the high-dose TFL group(all P<0.05).Conclusion TFL exerts a strong inhibitory effect on 1,2-dimethylhydrazine-induced colorectal precancerous lesion in rats,especially TFL with high dose.NF-кBp65 might be the key facto
