摘  要:目的 探讨miR-182-5p靶向原癌基因Bcl-2对骨肉瘤细胞增殖和迁移的调节作用。方法 收集2017年10月—2018年5月我院经病理检查确诊为骨肉瘤的组织及癌旁正常组织,检测miR-182-5p和Bcl-2在人骨肉瘤组织中表达情况。将骨肉瘤细胞株MG-63分为实验组、对照组和空白组;实验组将miR-182-5p过表达后转染到骨肉瘤细胞,对照组不做处理,空白组加入同等剂量的Bcl-2空白转染液。采用蛋白免疫印迹实验检测骨肉瘤细胞株MG-63中miR-182-5p调控Bcl-2的表达情况,细胞划痕实验检测miR-182-5p对骨肉瘤细胞株MG-63迁移能力的影响,MTT实验检测miR-182-5p对骨肉瘤细胞株MG-63增殖能力的影响。结果 人骨肉瘤组织中miR-182-5p表达水平低于癌旁正常组织( P <0.01)。过表达miR-182-5p后骨肉瘤细胞株MG-63中Bcl-2的表达水平相对上调。实验组miR-182-5p表达水平高于对照组和空白组,细胞增殖率低于空白组和对照组( P <0.05)。实验组骨肉瘤细胞株MG-63迁移距离低于空白组和对照组( P <0.05)。结论 miR-182-5p靶向原癌基因Bcl-2对骨肉瘤细胞增殖和迁移有一定的抑制作用,表明miR-182-5p在人骨肉瘤的发生发展过程中扮演着重要角色。Objective To investigate regulation effect of miR-182-5p targeting proto-oncogene B-cell lymphoma-2 (Bcl-2) on proliferation and migration of osteosarcoma cells. Methods Osteosarcoma tissues and adjacent normal tissues, which were detected by pathologic examination during October 2017 and May 2018, were collected, and expressions of miR-182-5p and Bcl-2 in human osteosarcoma tissues were detected. Osteosarcoma cell lines MG-63 were divided into experimental group, control group and blank group. In experimental group, overexpressed miR-182-5p was transfected to osteosarcoma cells, but control group did not receive treatment, while blank group was treated with same dose of blank transfection solution. Western immunoblotting was used to detect Bcl-2 expression regulating by miR-182-5p in osteosarcoma cell lines MG-63. Cell scratch test and MTT assay were used to detect effects of miR-182-5p on migration and proliferation abilities of osteosarcoma cell lines MG-63, respectively. Results The miR-182-5p expression in human osteosarcoma tissues was significantly lower than that in adjacent normal tissues ( P <0.01). The Bcl-2 expression in osteosarcoma cell lines MG-63 was relatively up-regulated after miR-182-5p overexpression. In experimental group, miR-182-5p expression was significantly higher, while cell multiplication rate was significantly lower than those in control and blank groups ( P <0.05). Migration distance of osteosarcoma cell lines MG-63 in experimental group was significantly shorter than those in control and blank groups ( P <0.05). Conclusion The miR-182-5p targeting proto-oncogene Bcl-2 has a certain inhibition effect on proliferation and migration of osteosarcoma cells, which indicates that miR-182-5p plays an important role in development of human osteosarcoma.
