摘　　要：目的:观察Neuritin对蛛网膜下腔出血后(SAH)神经元细胞凋亡相关的凋亡因子Caspase-8表达的影响。方法:雄性Sprague–Dawley大鼠分为四组,分别为假手术(Sham)组、SAH组、SAH+溶媒组、SAH+Neuritin组。采用视交叉前池注血法制作蛛网膜下腔出血的动物模型;通过苏木精-伊红染色法(H&E)鉴定SAH模型,Westernblot技术检测Caspase-8相关蛋白的表达,TUNEL法检测神经元细胞凋亡的百分率。结果:与SAH组相比,SAH+Neuritin组神经行为学评分升高(P<0.05),神经元细胞凋亡百分率降低(P<0.05),脑皮质神经元细胞内Caspase-8蛋白表达量水平降低(P<0.05)。结论:Neuritin可以下调凋亡相关因子Caspase-8表达,同时减少大鼠蛛网膜下腔出血后皮质神经元细胞的凋亡。Objective:To analyze the effect of Neuritin on the expression of apoptosis factor Caspase-8 in neuronal apoptosis after subarachnoid hemorrhage(SAH).Methods:Male Sprague–Dawley rats were divided into four groups:sham group,SAH group,SAH+vehicle group,and SAH+Neuritin group.The experimental SAH model was anterior cistern infusion SAH model.We observed the model of SAH by hemotoxylin and eosin(H&E)staining,evaluated Caspase-8 by Western blot analysis,and the percentage of neuronal apoptosis was detected by TUNEL.Results:Compared with the SAH group,the neurobehavioral scores of the drug-administered group were increased(P<0.05),the percentage of neuronal apoptosis was decreased(P<0.05),and the level of Caspase-8 protein expression in the cells of the cerebral cortex was decreased(P<0.05).Conclusion:Neuritin may down-regulate the expression of apoptosis-related factor Caspase-8 and reduce the apoptosis of cortical neurons after subarachnoid hemorrhage in rats.
