参慈胶囊联合顺铂对裸鼠A549肺癌组织DEC1及cyclinD1表达的影响

摘　　要：目的观察参慈胶囊联合顺铂对裸鼠A549肺癌组织分化型胚胎软骨发育基因(DEC1)及细胞周期蛋白(cyclin)D1表达的影响及其作用机制。方法培养人肺腺癌细胞A549并对裸鼠进行造模,筛选瘤体积(0.125±0.012)cm3的荷瘤裸鼠32只,随机分为对照组、参慈胶囊组、顺铂组、参慈胶囊+顺铂组,分别予以生理盐水、参慈胶囊免煎颗粒灌胃及顺铂腹腔注射等连续干预3w,测量体积后取材,采用免疫组织化学法、实时荧光定量(RT-PCR)技术检测cyclinD1和DEC1mRNA表达。结果参慈胶囊组瘤体积、cyclinD1和DEC1mRNA表达与对照组差异无统计学意义(P>0.05),顺铂组及参慈胶囊+顺铂组与对照组、参慈胶囊组差异有统计学意义(P<0.05),参慈胶囊+顺铂组与顺铂组差异有统计学意义(P<0.05)。结论参慈胶囊联合顺铂化疗能够提高疗效,并能降低cyclinD1的表达及提高DEC1mRNA表达。ObjectiveTo observe the effect of Shen Ci capsule combined with cisplatin on the expressions of DEC1 and cyclinD1 in nude mice with A549 lung cancer. MethodsHuman lung adenocarcinoma cell A549 was cultured and used in nude mice. 32 tumor bearing nude mice with a volume of (0.125±0.012)cm 3 were selected and randomly divided into control, Shen Ci capsule, cisplatin, Shen Ci capsule + cisplatin groups. They were given normal saline, herbal medicine, saline, herbal medicine and intraperitoneal injection of normal saline, saline, cisplatin and cisplatin respectively. After three weeks, the tumor volume was measured. Immunohistochemistry and RT-PCR were used to detect the expressions of cyclinD1 and DEC1 mRNA in the tumor. ResultsThe tumor volume, cyclinD1 and DEC1 mRNA expressions in Shen Ci capsule group had no significant differences compared with those in control group ( P >0.05);the above indexes of cisplatin and Shen Ci capsule + cisplatin groups were statistically different compared with those in control and Shen Ci capsule groups ( P <0.05). The indexes of Shen Ci capsule + cisplatin group had statistically different compared with those in cisplatin group ( P <0.05). ConclusionsShen Ci capsule combined with cisplatin chemotherapy could improve the curative effect, and its mechanism might be related to lowering the expression of cyclinD1 and enhancing the expression of DEC1 mRNA.
