摘  要:目的:探讨miR-423-5p在ConA诱导的小鼠急性肝衰竭模型中的功能。方法:制备ConA诱导的小鼠急性肝衰竭模型。RT-qPCR检测0h,6h,12h,24h小鼠肝组织和血清中miR-423-5p的表达情况,HE染色观察肝组织病理学改变,全自动生化分析仪检测血清ALT和AST。结果:小鼠肝组织和血清miR-423-5p表达水平随着肝衰竭进程显著性升高。小鼠肝组织和血清miR-423-5p表达水平与肝细胞坏死率、血清ALT、AST水平呈显著性正相关。结论:miR-423-5p可能作为反映肝衰竭严重程度的分子标志物。Aim: to study the function of miR-423-5p in the Con A-induced acute hepatic failure model in mice. Method: Liver failure in mice was established by intravenously injection with Con A. RT-qPCR was performed to detect the levels of hepatic and serum miR-423-5p of the mice exposed to Con A for 6 h, 12 h and 24 h. The hepatic histological morphological damage was detected with HE staining. Serum ALT and AST levels were determined using automatic analyzer. Result: The levels of hepatic and serum miR-423-5p increased significantly with the development of liver failure. The levels of hepatic and serum miR-423-5p had significant positive correlation with hepatic necrosis rate, serum ALT and AST. Conclusion: miR-423-5p may serve as a molecular biomarker for the acute liver failure.
