摘  要:信号转导与转录激活因子3(STAT3)是介导细胞内信号转导的经典转录因子,主要通过细胞因子受体/Janus激酶(JAK)/STAT3信号通路在细胞增殖、凋亡中发挥多种生理学功能。研究表明,异常活化的STAT3信号通路参与宫颈癌发生发展的各个阶段,并通过促进肿瘤微环境的形成,维持肿瘤细胞干性等促进宫颈癌的侵袭、转移和增强耐药性。采用多手段降低STAT3活性可显著抑制肿瘤细胞的恶性生物学行为,提示STAT3可作为宫颈癌潜在的治疗靶点。本文就STAT3信号通路的组成、调控特点及在宫颈癌发生发展中的作用机制、靶向肿瘤治疗等进行综述,以期对宫颈癌的临床治疗提供新思路。Signal transducer and transcriptional activating factor 3 (STAT3) is a classical transcription factor that mediates intracellular signal transduction.It mainly plays a variety of physiological functions in cell proliferation and apoptosis through cytokine receptor/Janus kinase (JAK)/STAT3 signal pathway.Studies have shown that abnormally activated STAT3 signaling pathway is involved in the invasion,metastasis and drug resistance of cervical cancer by promoting the formation of tumor microenvironment,maintaining the tumor stem cells.Reducing the activity of STAT3 in various means can significantly inhibit the malignant behavior of tumor cells,suggesting that STAT3 can be used as a potential therapeutic target for cervical cancer.This article reviews the composition and regulatory characteristics of STAT3 signaling pathway,its mechanism of action in the occurrence and development of cervical cancer,and aims to provide new ideas for the clinical treatment of cervical cancer.
