滤纸干血片保存方式对产前筛查标志物检测结果的影响

摘　　要：目的:探讨产前筛查滤纸干血片标本保存时间和保存条件对AFP、Freeβ-HCG检测结果的影响。方法:采集50例孕妇血样,每例均制备两份滤纸干血片标本,每一份上面包括3个血斑。一组2℃~8℃冷藏;另一组室温20℃~25℃保存。分别在保存第2d、7d、14d时,用TALENT-Ⅱ半自动时间分辨荧光免疫分析仪检测AFP、Freeβ-HCG。结果:室温组和冷藏组在2d、7d、14dAFP和Freeβ-HCG检测结果比较,差异无统计学意义,AFP和Freeβ-HCG冷藏组在2d、7d、14d的结果差异无统计学意义。AFP和Freeβ-HCG室温组在2d、7d、14d的结果差异无统计学意义。结论:滤纸干血片标本在冷藏2℃~8℃、室温20℃~25℃均可稳定保存至14d。开展筛查时应注重标本保存条件和运输时间,确保标本质量和筛查结果的准确性。Objective: To investigate the effects of preservation time and conditions on AFP and Free β-HCG results of dried blood spot (DBS) from prenatal screening. Methods: Blood samples were collected from 50 pregnant women, and two dried blood samples which contained three blood spots were prepared in each case. One group was refrigerated at 2℃~8℃(refrigerated group),another group was stored at 20℃~25℃(room temperature group). AFP and Free β-HCG were detected by TALENT-II semi-automatic time-resolved fluorescence immunoassay on the 2nd, 7th and 14th day. Results: There was no significant difference in the results of AFP and Free β-HCG on the 2nd, 7th and 14th day between room temperature group and refrigerated group. Within the refrigerated group and room temperature group, there was no significant difference in the results of AFP and Free β-HCG at 2days, 7days and 14days. Conclusions: DBS sample can be stored stably for 14 days at 2℃~8℃ and 20℃~25℃. The preservation condition and transportation time of specimens should be paid attention to ensure the quality of specimens and the accuracy of screening results.
