摘 要:目的检测配对盒基因-5(paired box,PAX-5)在Barrett食管、食管腺癌组织和食管腺癌细胞系中的相对表达,及其对食管腺癌细胞转移的作用和机制。方法以食管腺癌细胞、正常食管组织、Barrett食管和食管腺癌组织为研究对象,采用实时荧光定量PCR(qRT-PCR)检测PAX-5mRNA在食管腺癌细胞系中表达;qRT-PCR和免疫蛋白印迹实验验证PAX-5 mRNA和蛋白在组织中的相对表达。采用表达最低的OE33为研究对象,分别转染pcDNA3.1(+)-Flag-PAX-5质粒(实验组)和pcDNA3.1(+)-Flag-Vector质粒(对照组)至OE33细胞系,利用Transwell迁移和侵袭实验检测PAX-5的迁移和侵袭的影响,并用qRT-PCR和免疫蛋白印迹验证PAX-5基因迁移和侵袭的分子机制。结果食管腺癌中PAX-5mRNA和蛋白的表达显著低于Barrett食管和正常食管组织(P<0.001),PAX-5mRNA在食管腺癌细胞系中表达下调(P<0.001);Transwell实验研究显示:与对照组相比,实验组迁移和侵袭能力明显受到抑制;过表达PAX-5抑制上皮-间质转化(EMT)相关基因的表达(P<0.001)。结论PAX-5在食管腺癌中表达下调,可能作为潜在的抑癌基因参与食管腺癌的进程。Objective To investigate the expression of PAX-5 in human esophageal adenocarcinoma cell lines and tissues,and the effects and mechanisms of PAX-5 on migration and invasion of human esophageal adenocarcinoma cell lines OE33. Methods Esophageal adenocarcinoma cell lines,normal esophagus,Barrett’s esophagus and esophageal adenocarcinoma tissues were collected.Semi-quantitative real-time polymerase chain reaction (qRT-PCR) and Western blotting were used to analyze the relative expressions of PAX-5 mRNA and protein in these tissues and cell lines.PAX-5 was transfected into low expression adenocarcinoma cell OE33 cells,the migration and invasion function of PAX-5 were investigated,Western blotting and qRT-PCR were used to explore the mechanisms of PAX-5 in migration and invasion. Results Compared with Barrett’s esophagus and normal esophagus tissues,the expressions of PAX-5 mRNA and protein were lower in human esophageal adenocarcinoma ( P <0.001);the expression of PAX-5 mRNA was down-regulated in esophageal adenocarcinoma cell lines ( P <0.001);Transwell assay showed that compared with the control group,the migration and invasiveness of the experimental group were significantly inhibited,and overexpression of PAX-5 inhibited the expression of genes related to epithelial-mesenchymal transformation (EMT)( P <0.001). Conclusion The down-regulation of PAX-5 expression in esophageal adenocarcinoma may be involved in the process of esophageal adenocarcinoma as a potential tumor suppressor gene.
