摘  要:为研究担子菌单核菌株单一性,通过形态与分子鉴定比对,进行食用菌的杂交育种及遗传研究。用单孢稀释法涂布孢子,挑选单孢菌株进行锁状结构观察,判定的"单核菌株"用A和B交配位点进行扩增分析。结果表明:(1)随机挑选的11个单孢菌株中,1个菌株具有明显的锁状结构,被判定为异核体,而剩下的10个菌株缺少该结构,被判定为单核体。(2)依据交配位点A和B设计的引物扩增结果表明3个"单核体"为非单一性菌株,有2种A交配位点,但只有一种B交配位点。根据锁状结构认定的部分"单核体"可能会混有相同A或B交配型的异源菌株,建议单核体的分离采用单孢分离法,并且要结合分子鉴定。To study the singleness of monokayon from basidiomycetes, the morphology and molecular identification were compared for providing reference for hybrid breeding and genetic research of edible fungi.The single-spore dilution method was used to paint spores, and the single-spore strain was selected to observe clamp connection structure. The decidable "monokayon" strain was further analyzed with the amplification of A and B mating loci. The results showed that:(1) in the 11 single-spore strains, one strain had obvious clamp connection structure and was identified as dikaryon, while the other 10 strains were identified as monokaryos because they lacked clamp connection structures;(2) based on the A and B mating loci, the amplified results showed that 3 non-singleness "monokaryos" were non-single strains, they had two A mating loci, but one B mating locus. The "monokaryos" which are identified by the clamp connection structure is probably mixed with heterogenous strain carrying the identical A or B mating loci, thus it is suggested that monokaryotic strain should be isolated by using the single-spore isolation method combined with molecular identification.
